**Votes Needed** AGAR TRANSFER POLL!!!**Votes Needed**
    #17294521 - 11/27/12 11:30 PM (12 years, 1 month ago)

Just curious to see how people do things.
When making a transfer to a new dish, how do you place the wedge? Mycellium facing up, down or sideways?
You may choose only one
When making a transfer do you prefer to cut triangles or squares?
You may choose only one
When making multiple transfers, do you flame sterilize between transfers from the same dish?
You may choose only one
Was this a descent poll?
You may choose only one

There are reasons for placing the tranfers in certain ways. If It is a transfer from a dirt plate, I place the mycelium facing down just in case to help keep the bad stuff trapped. This is usally done with antibiotic agar. I cut in both squares and triangles depending on if I like the growth or not. Not that it changes the over all outcome but it does change the rate of the initial new growth. I dont sterilize between transfers off the same dish. I mainly flip the over just because of the way I stab them. The mycelium doesnt only grow on top of the agar. Its in all of it

Stop experimenting half way through your first grow. Grow it to maturity, watch it, learn from it. Do this a few times then experiment with different ideas and figure out what works best for you.

I don't sterilize between transfers from the same dish either.

But I do have a couple dishes where the wedge was placed facing down over a week ago and they show no growth period.

This is what made me start this poll.

I chose "flame sterilize" to mean "use a new pre-sterilized tool"

I, normally, place the transfer facing up or on its side.
It just depends on how the cut is.
I don't cut in squares or triangles. I cut in rectangles. :shrug2:
I don't flame between transfer from the same dish.
I get free sterile blades from another shroomery member, so I normally just use a new sterile blade between dishes.

& sorry, it wasn't a decent poll, because no STAL option was available.

It's an old OTD/Pubber.
I'm not sure exactly how he managed to get into every poll, but it's not a legit poll on here unless there's the option to STAL.

Now some agar pictures.
Pin on agar (after transfer):

What is the fastest way of colonizing your grain jars ?

Is this the right order :

fastest

1.g2g
2.agar piece
...
spore syringe
spore print
...

slowest

Well G2G doesn't really fit into the group... You would use a spore syringe or and agar piece"wedge" to inoculate your grains, then when your master jars are fully colonized you g2g those out... You can't really start w g2g or compare speeds with agar wedges...

That being said, g2g is by far the fastest way to expand your myc... I usually inoculate with an agar wedge then g2g the master jars out when they are fully colonized.... I've never done spore syringe straight to grain, since I like agar, and your forgot LC to grains, which is probably the fastest, but sometimes LC's can get you in trouble if your not careful....

Instead of triangles I cut medial triambic icosahedron's, face up, I flame sterilize between every cut. Awesome poll.
