this is the 3rd transfer after a liquid culture... its pretty consistent all the way around...

I feel like that grey patch below is dissecting.. but it's always difficult to tell in photos. Is that a shadow? Also, why are you taking such large pieces of donor tissue?

I'm no expert but I think I see a couple sectors for sure, maybe more. Better photo and lighting would help. Three transfers does not seem like many to me, especially if your lc was made from spores. Even from mushroom tissue most of my attempts at isolation took at least 4 transfers, though that just might be a testament to my noobness.

try taking a piece the size of a grain of rice, that way you're transferring less genetics

By the 6th transfer you may be done. I see very obvious sectoring at 10 2 4 and 6 o'clock.

The size of the donor tissue may appear bigger than it actually is because the dishes are a like 1/3 the avg size of a conventional agar dish.
yeah there is a grey shadow, its not myc or contam..
I have a stack of like 20 dishes right now going, some from agar some from grain, and a couple from h2o injected into the Myc-here is a pic of another dish I've got running..

That mycelium in the first pic is awful cottony. What species? If it is a cube, then I would look elsewhere. If it is a pan, or shiitake then it looks okay
-------------------- Stop experimenting half way through your first grow. Grow it to maturity, watch it, learn from it. Do this a few times then experiment with different ideas and figure out what works best for you.

Yeah, it is more cottony than what I care for, but I figure its the meduim, as it has some added bacterial inhibitors. It seems I get REALLY rizomorphic growth when transferring out of agar, if I take the super rizomorphic stuff back into agar it does this exact thing.. So I figure its just the growing medium, but what do I know -lol- .. Also, I looked at it under a scope, and I see clamped septa so thats a good thing, maybe I have alot more transfers do eh??

Always take the youngest myc from the outer edge

If your myc is not rhizomorphic, it may very likely be a sign that you are using too much dextrose in your medium. Take only half of the amount of dextrose when making agar medium next time and see whether that makes the mycelium look better.

Looks good, got that one nice rhizo strain Comin in strong at 10 o'clock... Next time try to just get one small little piece transferred like the size of a grain of rice or slightly larger, that way it'll start from the middle and grow nice and even outward.... Looks like you might have dropped 2 little pieces on your new plate, making it grow from 2 separate starting points...
