Non-Germination
    #1727394 - 07/18/03 11:05 AM (20 years, 8 months ago)

Hypothetically..

Some PF jars were prepared on July 7th. After 5 days at 80-90 deg F (trying to keep the temp => 85), there were no signs of growth. Finally, after 8 days, mycellium begins to appear, but: the mycelium is only apparent beneath one innoculation point. It would now be the 11th day, with no observable change in this trend. All four points on all the jars were equally innoculated pretty liberally with spores. Other jars are showing aggressive colonization at three of their four innoculation points. There seems to be wide gaps in colonization.

If this scenario were to occur to someone, it would be a symptom of what problem? What would cause this problem? Will the jars be able to fully colonize in spite of this unfortunate situation? Will reduced fruits be experienced? Can the situation be remedied?

some bacterial contams stop mycellium growth...give it a few more days and see if anything occurs.

Do the jars smell? How long and how did you sterilize them?

lemme guess...the jar with only 1 point was the first jar you innocculated right...sometimes the spores don't get evenly distributed around the solution in the syringe even if you shake the hell out of it...so the first few shots may be just water with no spores in it...the jar will colonize just fine but it'll take longer for the whole jar to be colonized...it shouldn't affect the fruiting...if you still have the syringe and didn't tamper with it after innoculation you could sterilize the needle and suck in some sterile water...often there are spores that stick to the sides of the syringe which you could use to innoculate more jars...

it's normal especially if you don't shake and/or the syringe is weak. afoaf ordered some texas syringes from a reputable vendor and three out of twelve pint jars of WBS have not taken even after three weeks. two of the nine that grew took nearly three times as long to begin germinating.

Hypothetically, for the sake of discussion..

Some of the jars were giving off a pretty vicious odor, so I evacuated them to the garbage can. I sterilized in a large pot, towel on the bottom, water about half way up the side of the jars, boiled for 1 hour.

GestaltAssault -- That's possible. I gave the syringes a good shake before removing the gaurds. I did a batch of EQ and a batch of B+. The EQ jars (3) are each colonizing at only one point. All but one of the (4) B+ jars failed. The surviving B+ jar is colonizing very well at three points, but there is a wide space that has me very concerned.

The three failed B+ jars were cleaned, refilled with substrate, sterilized using the mentioned technique and innoculated again on July 17th. Some additional Oaxaca jars were made on the 14th; no signs of growth as of yet. Too early to tell, I suppose.

>Some of the jars were giving off a pretty vicious odor,

Bacterial contamination, likely due to insufficient sterilization. Boil for longer, 90-120 minutes.
