If you're doing agar work, you always transfer from the most rhizomorphic growth, whether it's pronounced or not. That applies to first timers or agar vets :shrug:

If you're only making 2-3 transfers, you are close to isolates but they won't be perfect...myself and others (including RR) have found that sectoring for cubes doesn't even show up until after the 4th or 5th transfer.
