Is this normal growth for Oudemansiella canarii?
    #17083968 - 10/23/12 08:08 AM (11 years, 4 months ago)

I don't think so. This is after only 2 days:

Ignore the stuff towards the edge of the plate -- just camera reflections. What I'm wondering about is the very fast growing outer ring of linear mycelium, and the slower growing, fuzzy mycelium at the center. The outer ring is definitely mycelium, as it is only made up of radial threads.

This is from a rehydrated in sterile distilled water, dried mycelium sample, so there's a good chance of contamination. I've grown this from another dried sample in the same batch, with the same results.

I'm thinking that the outer, fast-growing mycelium is a fungal contaminant. If so, do I have any chance of isolating the good mycelium? And how? I'm thinking my only chance is to soak the thin sample in H2O2, with the hope of killing off a surface contaminating fungus. But maybe superfast agar growth is correct? I really don't know what normal mycelial growth is for this species.

Thanks,
Terry

Terry,
I can't say anything about the species, but in several fast growing types I have seen similar things.  Usually I have to hold the plate just so in the light to see the mycelium at the outer edge growing flat against the surface of the agar which is clearly differentiated from the aerial mycelium which grows next.  The species you are working with seems to have particularly obvious signs of this- I would not have been able to photograph the examples I am thinking of.

That being said, I hadn't considered the possibility of the fungal contaminants, as the only fungal contaminants I've ever seen are molds and they are obvious when they change colors.  If its a fungal contaminant like you suggested I may have several contaminated cultures and just not be aware of it.

That's a great point, laughingsol. The fungal contaminants I'm familiar with seem to all have funky, distinctive mycelium patterns or colors.

A few people here live in the tropics, and have cultivated this species. I'd like them to weight in on if they've seen O. canarii grow this fast. And this was at 70 degrees F (21 degrees C), not even tropical temperatures!

Hey Terry,

I was happy to see BP pop in on this one.  As usual, you are far ahead
on the new species efforts.  Kudos.

I had to look this species up, and though I am sure that you already
found this paper, it is species specific, and so I wanted to confirm:

Good luck!
