Spore Print to Syringe Question
#17000471 - 10/09/12 07:08 PM (12 years, 3 months ago) |

So I made a spore print off of one of my large caps of B+ onto tin foil. After it dried I scraped it into a shot glass filled with half a syringe. I scraped off the entire print into the sterile water.
Heres my question, the spore syringe I bought from spore depot had a few seeable specks of spores, which grew fine. Well the syringes I made off a whole print each are dark purple (TONS of spores in them).
How will this effect my grow with these? I did search but came up with nothing.
Heres my guesses.....
Colonization will be faster..... There will be an increase chance of contamination since there are more spores to grow bacteria on.....
Any insight?

if the contents of the empty syringe are sterile you should be fine, but i wouldnt bet any money that itll turn out well, why not sterilize the syringe?

Yeah, when you use your very dark new syringe you will only need a tiny little drop per hole as there are so many spores. You could have made more than 10 syringes from that print at the same time. The disadvantage of being so concentrated is the spores will germinate in many places and will compete for a while until they settle down (and thus potentially slow down colonization). If there's no contams in the jars there are no contams... having more or less mushroom spores won't change this. Hope this answers your questions.

tropicalfrenzy I think you answered my question...so should I dilute the syringes before I inoculate my next jars? Maybe do one jar concentrated and 1 jar diluted to see the difference under the same conditions?
and montock I know I wasn't very clear but my sterilization technique is pretty good(not perfect), I sterilize everything top to bottom including the room I work in. I'm one of those people who understand the difference between clean, aseptic and sterile.
Thanks

did you wipe the shot glass with alcohol first? you don't have to expel the water before adding spores. you should add spores first before expelling water. you should do this under a still air box or a decently sterile counter top where the air is still . turn the shot glass over after wiping it. sometimes i run a little water around the rim of the glass and pour it out to get residue alcohol that may be in the shot glass. its hard to say from your description if your syringe was made sterile.

If you can create sterile conditions you could dilute the syringe down (use sterile water too of course), but this is creating another risk point to contamination. Better off just using a tiny amount when inoculating. Feel free to do the experiment you suggested though, so you can see for yourself.

I may do 3 jars concentrated and 3 jars diluted to see, if i remember ill take some pics a long the way and post them for future if someone else finds it useful

It'll be interesting. Great.
I make my syringes so clear you can't see anything in them and they always work well. Have only read about using dark syringes and the potential ill effects, and the reason why, from much more experienced growers.
Please post pics

Just updating, did not get a chance to take pics, I inoculated/labeled 6 jars 6 days ago. I inoculated them from 2 different syringes that were made from 2 different spore prints, I did not dilute the syringes at all(both were very dark purple), however just for the sake of curiousity I inoculated 2 jars with a drop or two per hole, 2 jars with 0.25cc per hole, 1 jar with 0.5cc per hole, and 1 jar with 1cc per hole.
They are all being stored in the same way, fully enclosed box with a heating pad wrapped around the box on a low level heat (makes inside the box 80F+-.
All jars show some sort of growth so far. The jars with the most solution put into them show the most initial growth. The jars with a couple of drops have very little growth (maybe totally less than the size of a dime), and the jar with 1cc per hole has myc growth over 1/6-1/4 of the surface towards the inside of the jar.
All myc looks healthy too, pretty neat!

Unless its verry cold in with ur jars u don't need to incubate them room temp and ambient lighting do them good also what pf tech u using do your jars have holes in the lids with tin foil off?

Brf tek in 1/2 pint ball jars, I will get some pics up I'm the next couple days, I have 4 holes in each jar taped off, and lids on tight. I have read the whole arguement of fresh air to jars but my first time round turned out fine. If my myc stops growing ill adjust as needed. I'm in no super rush.
This is only my second time innoculating a set of jars, but I naturally love science, so running a small test of my own is fun. I read all the huge debates about how much to inoculate with etc. There's never one good answer lol, so ill see what works best for my set up...its not like I do.t have an endless supply of spores each grow

All of these pics are after 6 days of inoculation. I took pictures of the sides that show the most growth of each jar
Couple drops per hole

Couple Drops per hole

1cc split between all 4 holes

1cc split between all 4 holes

2cc split between all 4 holes

4cc between all 4 holes

Thanks, when the larger dose jars germinated was the myc more spread already? Seeing what is happening here makes me think that the greater amount of spore solution created many more germination points because of the sheer volume of liquid that there was to spread the spores around. This fits with the competing colonies theory, but I don't see the disadvantage yet. We'll have to wait ad see if they deathmatch for consolidation, but I really don't see that happening. 4cc is probably too much moisture to add, but other than that. Well, you have proved you could make your syringes go a hell of a lot further and save all that lab work making them.

Ok so they finally fully colonized and I birthed them into a FC.
There did not seem to be a deathmatch in the colonization stages, all went pretty evenly.
The 4cc cake fruited first, all fruits look average with the exception of one fat ass...next to the 4cc cake is a cake with a drop per hole and its the same result so far, gonna have one super fatty with a bunch of regular. The others cakes are producing good also, but nothing overly amazing. I did build a new/smaller FC for this grow so I had to play and adjust early on for the right settings which may have had a little effect.
Honestly I'd say spore amount has very little do to with growth rate for fruiting and colonizing. Water content, air supply, moisture, temp, and sunlight all seemed to have a bigger impact for me.
If for some reason I get waaayyyy mote fruits out of one cake than another ill post it, otherwise this is my conclusion.
Sorry for no more pics along the way, I could really sick for a few weeks and it wasn't a priorety lol
