Pin in myc water, safe to use?
    #16846260 - 09/14/12 12:30 PM (12 years, 4 months ago)

SO I took a glc a long time ago. Knocked jars they went fine fruited nicely.

Took the last half of GLC and injected it into a pint jar filled with sterile water to expand it. Knocked jars and went through a fruit cycle just fine no trouble. Saved the last 10-15ml of this GLC expansion in the fridge, just in case lol.

Tossed this in the fridge for the past ~8 months. I took it out of the fridge because I completely ran out of lc and I hate spores lol

I ended up not getting to my project and this lc sat in normal room temp for about 2 days. At the two day mark, I noticed very fast growing and thick threads of myc, spreading through the plain sterile water. Thought it was interesting so I let it be and set it back at room temp

Come day 5 from being out of the fridge I see a pin! it was growing right on the water, not on a mat of myc.

Is this still clean lc? In theory wouldn't the pin be sterile since whats in the jar has been tested and not contamed? Cause i would love to drop a sterile pin onto agar in front of my hood lol

Heard that if an lc pins it means its contammed though

Only one way to really know - put the lc to the test.  Pinning in subpar conditions can often be a signal of contams, but it may also mean that you have a great in vitro strain growing there.
