Strain Isolation on Agar from Colonized Grain?

I had 20 Agar dishes prepped, but discovered my spore syringes were too old. Nothing has grown from the past 20 plates I made.

Anyhow, I have a colonized quart of WBS that I used to drop 3-4 seeds onto each plate instead. Flow Hood / Flame Sterilized / YadaYada

Obviously this is not the best way to grow mycelium on Agar for strain isolation. I just figured why not try if the plates are going to go to waste anyhow. This was also a slow colonizing jar, so I thought instead of G2G, I'd make use of the agar plates and try isolation.

Figured I'd ask on here if anyone had decided to go down this route.

im not 100% sure but i don't think you can isolate a single strain from a multispore inoculation.

I could be wrong, ive only just started playing with agar and strain isolation

There's a chance you'll have to transfer sectors out a few times but it'll work for an isolate eventually.

Yeah, obviously I'd want to sector out. I'll want to do it sooner than later too if it works, so I can get rid of the actual grains.

Yeah, I am sure you will be able to separate sectors and possibly isolate something. Odds are that you have plenty of genetic material on that single grain from a multispore innoc. As for truly isolation a strain, that is a lot of work. Isolating a vigorous sector is probably what you are going for.

Stop experimenting half way through your first grow. Grow it to maturity, watch it, learn from it. Do this a few times then experiment with different ideas and figure out what works best for you.

Isolating from a pin clone may help save time in finding a fruiting strain also.

I just had one of my clone jars fruit and drop spores right back onto the agar. I'm kind of excited about sectoring out my "sterile spore print" lol

My first cloning experience was a success. Many after were failures. But I cloned a fat ass ps cyanofriscosas that I found in the bay area. I made a shit load of spawn and sprinkled it all over town. These are popping up everywhere now. Beginners luck. If i was a no such thing as coincidence guy, I would think that the mushroom  gods had blessed. Hell, I know they did.
