Cardboard cloning on substrate [Trying tek]
    #16817688 - 09/09/12 02:28 PM (12 years, 4 months ago)

So not sure if this one has been tried, or maybe any input or questions would be helpful. This is my first attempt at cultivation and with store bought mushrooms [portabello, and white buttons]. Wont be a easy task for someone who is just starting, but in my eyes i see it as a challenge and experiment. I plan on eventually working my way up to a professional level, and to take one bought syringe to be my only syringe after i learn how to clone effectively.

I have read about petri dish with agar, but as of right now i took a simple quick route. I have taken 3 mason jars, where i took anti bacterial soap to wash off jars, surface areas, and other materials before using. Immediately after washing the glasses they were put into the pot to steam/boil sterilize, with the cardboard under the messing to sterilize as well. This was done for 15mins of a increase in boiling and decrease to create different levels of heat in 3 stages. [Boil HOT, light boil, boil HOT] They were left in pot to cool in a decreasing steam and heat to cool. Taking out immediately after [glass and cardboard to dry while packing substrate] there cool enough to hold for long time in hand without burning, i packed  in the glass about half way [there small mason jars] with potting soil made of bark, sphagnum peat moss, and poultry litter [here. im not sure if poultry litter would work as well as cow manure. After packing i closed each jar, before beginning the cloning process, which then i took a sterile cutting knife which was washed and put under flame. to cut the mushroom in half. I took the sterile cardboard cut it into 1x1 inch pieces and took mycelium from different parts of the mushroom that were known to be most effective. Some were layers and some were chunks [i figured it increase my odds putting more variables in] which were added in layers. such as the diagram, with each SOLID line representing a piece of cardboard.

Each jar [3 jars made] had different amount of holes cause im trying to see which one grows more effectively, though its not exactly a great controlled experiment. Each was covered with a breathable bandaid over the holes. Jars amount of holes were 2 - 4 - 5

They were washed of any soil on the lip of glass, and or tops before being placed in a clear container in a mildly dark room. I put them in a small container to keep an extra chamber to make sure they stayed around 70degrees.

ohh.... they were sprayed one last time before they were stored, about 4-5sprays.

Glasses go on top of meshing, and cardboard underneath meshing

This was after boiling, so water was little low. It would normally go to the top of meshing or to the mouth [bottom of glass] so it would bubble into it, as well as steam it.

All of that is fail.
You need to do your research.
Find a tek to grow something you want to grow and start there. Follow directions to the letter.
Don't try to innovate. Trying to be a mega genius who hops into a complicated science based hobby and immediatly owns it and turns it upside down is well... complete fail
good luck

Want to clarify more, because ive read it is do able through cardboard tek, but your right i am trying to innovate. This was more of a experiment then an actual first start. Are you trying to say though stay strictly to the cardboard tek if im going to do it, or dont even bother with that tek?

dont even bother with that tek its fail
agar if you want to continue that path but you should make a legitimate start by buying a syringe and following directions
learning this hobby isnt easy

I plan on buying syringes. I picked up some stuff to do it by the book. I was just trying this tek, with a little innovation to see if i could do it. Ive watched plenty of movies and readings, and plan on doing it legitimately. I appreciate your input, because i was just curious if people though this would actually work to someone who has tried cardboard tek as well.

You can use cardboard in a pinch if you don't have agar.  But agar is the preferred medium. Ive used cardboard successfully for cloning pins til my agar showed up.  If you were going to use cardboard you could do this-

-With a sterile scalpel and using a still air box, cut a small piece out of the inside of the mushroom's stem and place it in your sterilized dish or jar

-Let it colonize the cardboard for a few days.

(not my picture, its from here)

-Cut a piece out of the colonized cardboard and transfer it to another sterile jar or dish containing cardboard.  Let that colonize. This second transfer is in order to "clean" the culture

-Prepare some grain jars and (yet again using SAB)transfer pieces of the colonized cardboard to the jars.  grains-> bulk sub-> fruit

Imo its better to let the mycelium colonize the cardboard and then refrigerate til you have agar.

Yeah so Ive read, and seen it was possible. If i get any growth, or anything for that matter, ill keep pictures posted on here.
