PC'ing LC medium directly in the syringe

It's been years since I cultivated, but I'm thinking of messing around again. When I last stopped, I was fooling with an idea to mix up a large batch of LC medium, suck it up into 100 cc syringes, PC en masse, then suck a little bit of culture up into each syringe, allowing the LC to grow directly in the syringes.

I couldn't make it work. The primary problem was the pressure increase within the syringe during PC (I assume due to the medium vaporizing) was causing the pistons to blow out of the syringes.

I was playing with the idea of building a rack that would hold the syringes and keep the pistons from blowing out. But there are other potential problems with the whole idea, and it could be that it won't work for some other reason I am not aware of.

Should I keep experimenting down this track? Has anybody tried this before?

sounds senseless to me. First you need pc'able syringes which do not melt.
Also why dont't you make a normal lc, put your culture in there and suck it up then?

If think those syringes are really prone to any contam hidden in the culture you want to suck up..

Another thing: How do you get the culture in the syringe into small pieces. In bigger batches you could use a magnet stirrer but here?

this are just the first thougts and I would say that you shouldnt go for it

You should make a big jar of GLC then suck that up into your sterile syringes. That way you:

1) Know you're not growing contams
2) Don't have to worry about exploding syringes

Its stupidly easy to make GLC and you don't even need a big pressure cooker. One quart jar will make enough to fill a few of your 100cc syringes.

You could always PC the syringes with the needle end pointing up...
