easiest way to an isolation

after i spawned a few jars i noticed there were myc chunks stuck to the side of the jar. i happened to do about 5 extra agar dishes than needed the day before so i thought, what the hell, i'll give it a shot.
i scraped the tiniest little piece imaginable from the jar and set it on the agar. i didn't think anything of it and sat it down, 6 days past when i saw the little booger hiding behind everything else so i decided to check it out. and boom. a bonofide mono culture, perfectly circular. not the desired rhizomorphic, but nonetheless, a monoculture.
what are the chances?

^^ what he said. If you started with MS, the chances of collecting one strain from a jar of thousands is unlikely. Fasts way to mono culture is a clone. Transfer a piece of the culture to a new plate and see what grows out, see if it segments. From weak spore solution it still takes me 4-5 transfers to isolate. Good luck.

clones contain just as many substrains as MS, the fastest way is transfers from the very edge of a sector off an agar dish until no more sectors grow out and you have an isolated strain of whatever your growing. 
cloning is best for choosing charactoristics that you want but it wont get you near a monoculture.

I have cloned and had a mono culture and I have cloned and had as many as 4 strains. Luck I suppose but it has been the fasts way to a viable mono that I have found. Certainly a good point.

It seems you learn something every day. I always thought that a cloned mushroom tissue gave a monoculture. And i also thought that if you have done a MS inoculation it was thousand of substrains from the beginning but that they made war and the stronger took over the weaker so the amount of substrains constantly become less and less until only one remained. This ofcause take long time.
So the substrains doesn't decrease in a mulispore inoculation with time? And that a single mushrooms contain many substrains... news to me.

Nearly every post above has factual errors in the poster's understanding.
Scraping the glass after dumping grains out of a multi-spore inoculated grain jar may or may not yield a monoculture. It has about the same chances of a fruit body being a monoculture.
If it is a single sector isolate, it really doesn't matter unless it is a stellar performer.
Often, clones will deliver a single sector isolate, but not always. Many strains can be active in a single fruit. By the process of anastomosis, most compatible strains combine into a single genetic organism. Those strains which are not compatible remain active in the mycelium, and actually participate in forming fruits through processes yet unknown to science. In other words we've observed it, but don't understand how it works. RR
