Dry tissue cloning/spore extraction ???
    #1650829 - 06/21/03 01:29 PM (14 years, 5 months ago)

I've read that it's possible to clone dried sclerotia by dipping tissue into a H20/H202 solution and I've also heard of people who have had sucess extracting gill tissue and placing it on gentamycin/benlate agar.

I've got some dried Boletus edulis and was wondering if anyone could give me more advice on this subject ? Do I need to rehydrate the tissue in any way before I dunk it in the H20/H202 ???

You don't have to rehydrate it - just place it on agar. I think that the tissue isn't alive anymore but you can hope that some spores will germinate. However Boletus edulis is a mycorhizzal mushrooms and it grows very very slow on agar even if you clone fresh tissue. Good luck - you'll need it!

Anno did this with Volvariella volvacea:

Thanks for the info zeronio, it sounds as though growth on agar would be near on impossible with dried gill tissue so I've decided to make a cepe, brandy and peppercorn cream sauce to accompany my steak tonight
