It is used only once. Its purpose is to tell how many microns are in each reticle division.
Once this is known you can write it down, allowing you to use the reticle (in the 10x eyepiece) to measure spores.

I also will tell you if it goes on top of a slide. No it doesn't. It is a slide actually. (See the photo of mine)

A reticle is a little plastic disc that goes into an eyepiece which has markings. You'll see a grid or measurement bar when you look through the eyepiece. Usually people order an eyepiece with the reticle already installed, though you can just get the plastic disc and install it into an existing eyepiece.
