Making a spore syringe???
    #16199492 - 05/08/12 11:34 AM (12 years, 8 months ago)

I have been growing mushrooms for a few months, but I am a complete noob at the whole syringe/print game. I have an idea for a simple, yet effective tek. Hopefully you guys here can tell me if I'm heading in the right direction, or what I should do instead.

First, I will select a mushroom that is of good size and has opened its cap, but hasn't release spores yet. I will cut the stem of the mushroom off as close to the cap as possible, without messing up the cap. I will use sterile scissors (wiped with rubbing alcohol).

Meanwhile, I will take an empty pint jar, and sterilize it in the oven at 250 Fahrenheit. I will use foil as a lid for easy access, this will also be sterilized in the oven.

I am going to thoroughly wash my hands before proceeding!

Once the jar is cooled, I will open the foil lid and place the mushroom cap inside the jar. I will re-cover it with the foil, and I will place it on a shelf for a few days.

After a few days, spores should have been released. I will use a sterilized knife (wiped with rubbing alcohol) to remove the mushroom cap from the jar by puncturing it. I will re-cover the jar with the foil to keep the spores safe.

I will boil some water, and I will fill some syringes (the same ones I used to start my projects in the first place, I am reusing them) with boiling water. This boiling water should sterilize the syringes.

Once the syringes are completely cooled, I will eject the majority of the cooled water of the syringe into the jar with the spores. Using a sterile tool, I will gently stir the jar, as to mix the spores with the water. I will fill the syringes with the water/spore solution.

This should be refrigerated and used for inoculation (I will be making cakes again) as soon as possible.

Does this sound like I am on the right track?

Sounds like most of what your doing is correct. You dont really need to wait a few days for a print though, they usually dropy in 24 hours or less. If your are brand new to printing and syringe work, I would suggest not trying to come up with your own new tek's and following some tried and true methods.  You can find an easy follow print/syringe guide here.

Good luck!

Your plan sounds good to me.

When you go to choose a cap for a print. Pick one that has almost flattened out completely. If you want a thick, dark print, let it sit for two days.  The cap will start to turn upwards, that's when you know it's good to go.

edit:  I also don't think it's necessary to sterilize the jar, but it's not harmful either.  I think with more experience you'll find that in the beginning we tend to over do the sterility of our projects.  Better safe than sorry I suppose.  But I never sterilize my jar, just wash with soap and water is good enough.

OP you got a great game plan, the only thing I do a little different from what you are describing here is, I put the water on to boil and let it boil for twenty minutes (killing any bacteria in the water) then suck up boiling water in the syringe and let it sit for 2 minutes. Then I squirt it into the sink and repeat three times. On the final draw leave the water in and allow to cool before I use. Otherwise your on the money.

Goodluck :fasted:

For my first time making a syringe I did pretty much what youre talking about and I was pleased with the results. I used a 1/2 pint jar for the print which was sterilized it with alcohol (no oven, pc, or steaming) and kept sealed with its self-sealing lid. I allowed approx. 8 hours for the print to build up and it was DARK. I used a sterile syringe which I made the night before and expelled the water onto the print while using the needle to break up the spores (only open the lid enough to get the syringe in there - do not remove it completely), then sucked them right back up. Everything worked great and the jars I injected with this syringe took off like crazy! Just make sure your syringe was sterilized properly (bowling water expelled into the sink a few times) and that your print taken in a sterile area.
