If the spores & sprinkler were clean then it could work, but you would have better chances with a syringe.

Well, it will most likely start to colonize, the question is if something else will colonize as well, ie mold or bacteria. In my experience grain is not workable with out hepa filtration of air in the incubation area. Of course, a big part of that depends on the environment one is growing in.

It works also without flowhood but you have to use peroxide and inoculate with agar wedges.

You don't need hepa filtration for incubation. You just need to be sure you do not introduce contams into the jar before sealing it. You must use a filter disc or something, but why would you need to filter the air for incubation? The jar is sealed. Contams can't penetrate glass.

Yes your jars will probably colonize, if you were clean about introducing the spores.

Though you should pressure cook for at least 60 minutes, 90 minutes is better.

What are you talking about calicyco? Surely if, as in the process that YellowPurpleHills described, you simply sprinkle in the spores in an open air environment, then filtering the air which will come into contact with the unprotected substrate after sterilisation will significantly reduce the risk of contamination?

I figured he had "sprinkled" via a syringe. I see your point if he had just "sprinkled" spores from some unsterile means, though I dont see any mention of open air.

Anywho... YellowPrupleHills.... spores work on rye, but as was posted above you do have to be very wary of contamination. Not after you have sealed the jars, but when you put the spores in of course.

I think the problem with using spores on rye is the extra time it takes to germinate. Rye is very contam prone. However if done sterile its not an issue in my opinion. I would be more worried about your PC time then the extra few days it takes for spores to germinate.

Also, spores aren't really "bad" just inefficient. With rye, its better to use mycellium of an isolated strain. This is purely for efficiency and using multispore will work, you just have to deal with many competing strains. It is usually assummed that if you are moving to rye, you are ready to move to single strain isolates, which have much higher yield potentials and grow much more uniformly.
