Safe to leave petri disc uncover in the glove box?

Hi all, Is it safe to leave the petri disc uncover in the glove box (after pouring in the agar) for the agar to cool down before covering them? The point is to prevent vapor collected under the petri disc's cover. I understand that 1 of the way is to wait for the agar to cool down enough before pouring them into the disc but its difficult to get the timing right and I always end up with vapor under the disc's cover and it obscure the view.

Its not really safe to ever leave 'em uncovered, or without sealing the sides up.(with parafilm/medical tape/ Saran wrap) but you could make the agar mix up, but leave out 1cc of water. And when your preparing your dishes, pour the mixture into the dishes, and spray a little diluted h2o2 into the dishes (hydrogen peroxide) like 3% h2o2 to water ratio. But the down side is no spores will germinate on the dishes after sprayed with h2o2, but already germinated spores will do fine on it (mycelium, LC, or any live tissue samples such as a clone or cutting on a mushroom.) Lastly, no contaminant's spores will germinate on the h2o2 unless it was put on the dish already germinated (on the culture you put on it.)

EDIT: you can check out my gallery, all the cardboard tek clones were prepared in a nonsterile environment, and i check on em in a nonsterile environment. (Notice nothing but healthy mycelium, thanks to the wonderful chemical h2o2, i spray a diluted h2o2 solution on it every time i open or close the lids, and while preparing em.)

Yup, don't let 'em sit uncovered, even in the glove box. I was having condensation problems, and placing a few dishes filled with hot water at the top of the stack cured it. Condensation happens when there's an abrupt temperature change, like at the top plate exposed to cooler air. Providing a path for this heat can solve it.

Hi Ninja69 & Terry M Thanks for the tips & comment. Will try the h2o2 the next round. 8 ) Just make myself a glove box and am trying it out.
If I have 3 small spot of contam underneath the agar (between the disc inner surface & the bottom of agar) but it seems to stop growing, shall I transfer the culture to another clean disc or do I just observe for a couple more days?

I would suggest transferring the cleanest portion of the culture as soon as possible. But do everything slow and medicals, when you open the dish fast the contam will sporulate, causing the whole dish + surrounding area to become contaminated.
Also, contams usually grow pretty fast compared to mycelium.

So could i use a mason jar with hot water on top, instead of a petri dish??

You were right. I should have transfer two days ago. Now my culture tissue started to have wet spot. I tried transferring the grown mycelium which is still very small amount. Hope it is clean and will grow.

Sorry bout your lost, but its all a growing experience learning new technics here and there. Just building on the basic's of being sterile. Hope you can transfer a viable specimen.
