Hi there,

About to make some spore prints, also for incoluations purposes. I have a cheap glove box, no HEPA filter, wondering do I need to spray a solution of 10% bleach 90% water in the air inside to first sanitise before making my prints inside the glove box? I really want to avoid contaminant and I ive in an old house (worried about air borne mouldy contams).

I also have like a 80% contamiation rate in my jars, whereby aroudn 50% -80% colonisation, I get a single contam somwhere most the time.

ANy suggestions? :smile:

Some vet growers have even said plain water do wonders. You just want some form of liquid on the walls so any spores will stick to it. I might about 25% iso alcohol with water in a spray bottle, probably not even needed though.

i never used bleach
h2o2 is what most people use
and all it does is help prevent contams
other then that there really is no need

and for you glove box use lysol to spray in
then wipe everything off with rubbing alcohol
let everything dry
then go to town

And then you light the torch to flame sterilize.  :blowupie:

There is no reason for anything but soap and water in a glovebox.  You're not dumping your spawn out on the floor of the thing, you're only using it to block wind currents.  Gloveboxes are not sterile and not meant to be.

when i used a GB i was extra cautious and thats how i did it
now i just innoc in open air
the only thing my GB is for now is agar or grain to grain

thanks for reply guys. Just setup the GB as per your suggestions and placed the caps down.

There's some sort of contradiction here (as per my previous opinion) as to why we don't sanitise in this part of the process as opposed to during inoculation. (I'm clearly wrong about this but given my 80% ish contam rate I'd like to know why)

If we make our prints in a non-sterile environment, can't airborne contams (which I was under the impression there were billions floating around in regular air, simply be floating around our GB and land on the spore prints, which would then find their way into the syringe, and accordingly into the substrate anyway?

I guess I can see it reduces the risk significantly as the h20 in the syringe has been PC'd so your only risk of exposure becomes the print... am I right?

Also how important is flame sterilising between inoc holes? My crappy 2ML syringe has been whipped into the garbage as its not airtight and was driving me crazy, I've got a super fat 1oz plastic flavour injector now, but I won't be able to flame it.

th GB that most people use and make really isnt a glove box
if im not mistaken a real glove box has the air sucked out and fresh filtered air put in

what your gonna make is really a SAB or settled air box
the point of this is that the air in the box isnt moving
when the air isnt moving it lets all the contams(mold spores and bacteria spores) sink and settle on the bottom

10% bleach is fine but so is pure water or lysol as the liquid will hold down any airborn dust and bacteria.

Even plain water will work. What you're going after is just some moisture on the walls of the GB or SAB so contams can stick to it and get pulled out of the air. Think of the same effect as how some jewelry stores will leave a cup of water in their window display. It keeps them from having to dust off the window display for a much longer period because alot of the dirt that floats around in the air gets pulled into the water.

No. You don't want water on the walls of your still air box.
Warm soapy water, rinse with clear water, air dry. <- That's how you clean a still air box.

sweet think I get it, gravity pushed the floating particles down inside the box and no currents means no contams flying around, cool :-)

That jewellery store reference, are you saying the slight negative charge of the water attracts the dust?

Yea it does, if you've seen it in window displays then you get what I'm talking about. It won't be pulling contams out of the air like crazy but it helps especially in enclosed/semi-enclosed areas since dust/contams will stick to the water. Essentially slowly pulling it out of the air and keeping it in the water.

You could but it's not needed. Why kill contams you can simply wash away?
Warm soapy water, rinse, air dry. That's how you clean a still air box.

(some of those wipes are alcohol based, watch for flames if you have that kind)
