Innoculating ...
    #1606445 - 06/04/03 12:13 AM (14 years, 1 month ago)

In most any teks, they tell you to "innoculate" but they dont say how. I'm a newbie, so bear with me. Do I remove the lid to innoculate, or innoculate through the filter (seemingly defeating the purpose of the filter) ... but if I were to remove the lid, wont contams get in?

Luckily, they tell you to inoculate.

Thats just my question though, lots of the teks tell you to have a filter in the lid, so wouldnt poking through that defeat its purpose? Or are they assuming you'd be innoculating in a completely sterile enviornment? In which case you'd innoculate and then put the filter in... I dont know, I'm confused.

fuck the filters. just make small holes in the lids of the jars presterilization that are just big enough to fit the needle through. then after you inoculate put tin foil back over the lids and toss them in the incubation chamber. works everytime.

yup, burntoutstarzzz has got it right. put a few holes (as recommended in many of the teks out there) and pressure cook with the foil tightly wrapped around the tops of the jars. then working in a sterile environment, and upstream from contams (oven works well), sterilize the tip of the syringe, do the innoculations, and cover again. wear gloves, wear a hat, wear a mask. don't sneeze. ;-) you'll know in a matter of days if you have contams. throw those out and work with the rest. good luck.

If you feel unsure about contams entering the jars through the inoculation holes, here's what you can do: Put some sticky tape over the inoculation holes when you're done inoculating. The drawback of this method is that no fresh air can enter the jar, which might stall mycelium growth. Therefore, pack the substrate mixture very lightly into the jars, and leave some open space (say, an inch) between the mix and the jar's lid. This should create a buffer which holds enough air for the mycelium to grow.

Happy growing!

You should drill a total of five holes in your lids. One in the center for your poly-fil and then four more on the outer edge of the lid.

Like this.

I tape the outer holes with clear packing tape, and put on the foil. Then once they are preasure cooked and cooled I just steralize the needle and the surface i am innoculating, let air dry, innoculate the jars through each of the small holes, re-tape, re-apply the foil, label the jars and thats that. I use my glovebox for this step. The safer you are about contams the better off, at least in my book.

Hope this helps

Peace

Sorry, my bad. Inoculate ... Didnt think one letter would be such a huge deal for all of you. I've seen much worse spelling out there.

Also, I havent been able to find vermiculite or BRF around so I did the honey tek. Incase that makes any difference. I didnt find out until after I tried it a couple times that it calls for ultra sterile conditions.
