4 days in and no sign of bacteria. I would suspect if there was any bacterial contaminates they would have shown themselves by now. I plan to leave the plates for another week or two just to see if any contaminate spores will germinate.
This is by no means concrete evidence, or an experiment with constant variables. I encourage others who have more background doing experiments and with science to perhaps carry out more research.
There is no argument that wiping down an instrument that has been flamed sterilized opens a vector of contamination. The question is how likely is that vector to ruin your projects. I will update this post if anything starts growing.

9 Days in and still no contaminate growth of any kind on any of the plates.
I plan to keep them around for another 7-10 days.

No contamination across the board. But remember this is a small test that did not follow strict standards...more testing would be needed to put it into stone.

So the experiment didn't prove anything?

It proved that a flame sterilized needle is clean. If the OP repeats the experiment, he should try alcohol without flame sterilization and watch the green and slime(bacteria) appear since alcohol doesn't sanitize the inside of the needle. RR

Maybe work with reused needles since a lot of people save them to use later for LC and stuff.
There could be a lot of variations to test.

Thanks RR for the explanation. I'm using a glass lamp with 70% isopropyl alcohol solution to sterilize my needles. Is there a better method?

If you are using that alcohol to fuel a flame that flame sterilizes your needle, then no, there isn't really a better method than that.

You should be using denatured alcohol in alcohol lamps. It's cheaper than iso, too.

Ty for putting the time to do something like this. i know you stated earlier that you didn't do a control dish. would be interesting to see a plate that was exposed in open air for a few sec. just to see the load in your lab. Again, ty for doing the experiment

Perhaps at a later time I may go into things a bit deeper and more structured, but for now I am far to busy.
Everyone please feel free to conduct your own experiments.

The hypothesis, and the point, is still a good one though
- A flame-sterilized whatever (scalpel, syringe, etc) is sterile. - Alcohol-soaked whatevers (swabs, paper towels, etc) are almost certainly not sterile - and the alcohol won't make them so. - Don't touch a sterile thing with a non-sterile thing; doing so will make it non-sterile, almost by definition.
Flame-sterilize, then go to work. You can't do better than sterile. Sterile is 100%.
Alcohol is certainly useful for sanitizing things, and you should definitely sanitize any relevant item you can't sterilize. You can't sterilize your gloved hands, for example, so we do the next best thing, and sanitize them with alcohol.
