I got my culture and have been busy making slants, plates, and spawn-- so far so good. If successful I'll be using the spawn to start bioremediation trials!

does anybody know the logistics/chemistry of infusing polyeurethane into agar? if I just melt it and add it to the mix will it dissolve?

Water based polyurethane, it must be added after sterilization. If you mix it in before pressurecooking it precipitates out into strings and chunks of plastic. I was able to confirm that there does not seem to be anything in the water based polyurethane that inhibits the fungal growth, though I did confirm that my strain will not grow on straight polyurethane agar alone.
The water based polyurethane is opaque, you can see the fungus eating it because it will clear up around the mycelium on the plate.
I will be continuing my experiments here soon, I will try sterilizing the polyurethane separately then adding it, if it can survive pressure cooking on its own.

Sorry I haven't been on lately & haven't received a culture yet... I had some members involved PM me on here with address confirmation, but I haven't been on to confirm because I'm getting ready to move to NC.
Hopefully there's still a little guy waiting for me *fingers crossed* I was looking forward to checking it out.

I think UV sterlization may be more ideal for sterlizing polyurethane, no?

I don't have a UV lamp yet.
And UV tends to only sterilize on the surface, though I do think it can be used on liquids. The water based polyurethane that I am using is opaque and would not let enough UV in to sterilize, though honestly I am not sure if the polyurethane would even be a contamination vector. Depending on the cleanliness of the factory it was made in and how careful you are about extracting it from the can, for some reason it says not to shake so I have not shook it since I opened it, in theory I should be able to pull clean polyurethane from beneath the surface, it also helps that you are only supposed to add a very small amount.
I am thinking about forgoing sterilization all together with the polyurethane and just adding it to some agar before I pour it. I just haven't had a batch of agar that wasn't already going towards another project lately.
I also need to get a hold of this culture from the co op buy. I am not sure that my personal culture has the plastic eating ability, though it could just be because I screwed my agar up the first time.

Slants are very hard to get into Australia due to customs restrictions. It would be best to send a very small agar wedge in an regular letter envelope, hopefully it won't be bumpy.
