Freezing Syringe/LC
    #15932969 - 03/11/12 11:58 AM (12 years, 10 months ago)

I have 3 syringes that I don't plan on using for at least a year (maybe longer). Would it be a good idea to freeze them for long term storage provided I let some air in? How about freezing liquid culture?
One more question, approximately how long are syringes viable for in the fridge?

Although I appreciate any comment, please don't guess. Only reply if you know the truth for a fact.

Thanks....PS- I did do a search but came up with mixed results as usual.

Looking for a trusted cultivator to chime in if possible.

What I meant was, say I decided to freeze a syringe and didn't let some air in to allow room for water expansion and the syringe cracked, well to me, freezing doesn't work. But, I still will have a doubt because I was not 100% sure I did it correctly because I didn't allow air for expansion and only have that 1 point of reference. Now what if someone asked that question and I said, no way, you can't freeze a syringe. Well, I'd be guessing because I only had 1 experience and didn't follow all the things I probably knew I should or I forgot a thing or 2. So I can't really say it doesn't work because I'd be guessing. I didn't really mean to suggest that people would make stuff up. I just meant that I would like to hear from someone with more than 1 experience or from someone who has 0 and heard it from a friend or read it.

I appreciate any and all help from anyone who is willing to share it.

Thanks....

Anyone have different experiences than Skuz?

Im pretty certain that freezing a spore syringe will kill the spores. I know they can survive over winter but that is without being trapped in ice.

My advice would be to just leave it in the fridge. I have left a syringe in the fridge for 1.5 years and it still worked fine.

Also if you are concerned about something keeping for YEARS, maybe make a spore print and vacuum seal it? It will last forever that way.

How about a vacuum sealed spore syringe? 2 out of the 3 are vacuum sealed, will that keep them viable for a longer time?

freezing tends to bust the cells walls of spores, and hyphae (myc)

Don't freeze your syringes. Here's why:

The outermost layer of a cell is called the cell membrane or plasma membrane. These terms mean the same thing. The membrane is made of phospholipids.

You can think of these as a balloon with two string. The 'phospho' would be the balloon and the 'lipid' would be the two strings. Lipid is a another word for fat, so the strings actually represent fatty acid chains (all fats are chain-like molecules)

Here is an animation of a cell membrane.

If you were to mix pure water with pure phospholipids and shake it up really well, you would find that over time, the lipids spontaneously attract to eachother and form small vesicles. The fact that this happens spontaneously (aka these shapes are the most energetically favorable outcomes) has been cited as evidence for the creation of life in the oceans of early earth. A recent study, published Jan2012 suggests that the first cells began in deep sea hydrothermal vents.

But continuing on, the forces that attract these lipid chains to eachother is very weak (think: frozen ice cream is easier to bite through than frozen water). So the reason that 30 degrees F will kill you while 36 degrees F won't is because when water freezes, it expands and ruptures your cell membrane. This expansion also likely denatures (renders useless) many proteins within the cell.

It should also be noted that bacteria and plant cells have an extra outer layer called a cell wall, which is rigid and made of complex sugars/proteins. The drug penicillin, which was derived from the mold Penicillum (a fungus!) in the early 20th century actually targets the protein in bacteria responsible for making the bacterial cell walls. Fungi-derived antibiotics is an abosolutely fascinating field of study and is the focus of Paul Stamets' latest research.

So to finish up, spores are cells (phospholipid bilayer, contains DNA) and will be damaged irreparably by freezing.

Don't freeze your spore syringes.

ha.. i just realized how much shit i posted. my bad...

Anyway, here's an even better animation. the blue green sea of craziness from 0:25 to 0:30 is a phospholipid bilayer.

thanks for letting me nerd out in ur thread  :strokebeard:
