should a mono culture be a mono culture after fruiting?

i started this several months ago and at the time did not have master culture slants. i had a mono culture from my understanding this is what it should look like . I have done a good bit of research and watched roger rabbits video on the subject. my reasoning is when you start from spores onto agar there a many different sectors available to you. the way to achieve a mono culture is to keep taking sectors of this growth until there is one uniform pattern of grown as shown above. my problem came about because as i started with a mono culture after it fruited and live tissue was transferred to agar there were multiple sectors in front of me. this is my question why was it not still a mono culture? does this have anything to do with cell degradation (from the myc traveling so far and splitting so many times) or do i just not understand the concept of a mono culture

A cloned fruit from an isolated single-sector strain will produce isolated single-sector fruits. A clone from a fruit grown on a multi-sector culture will often, but not always produce fruits with more than one strain active.
I wouldn't declare your petri dish above to be a monoculture. That's pretty thick growth. There could be hundreds of strains active there. RR

Wow that's really interesting. I was assuming that the dishes I had that looked like the above (except a little less tomentose, more rhizo looking) were mono cultures as well. So is there a reliable way to determine a mono culture with the naked eye?

Transfer tiny pieces of mycelium to new plates, putting two pieces from opposite ends of the original onto new plates about an inch or so apart. If they grow together and fuse without a 'seam', then they're the same strain. If they form a line of separation where they meet, they're non-compatible strains. RR

I thought I had mono cultures too, but apparently I really didn't have a clue. Thanks for sharing the insight about how to test for a monoculture. This explains... so much...
