liquid culture help!
    #1589263 - 05/29/03 05:26 PM (13 years, 6 months ago)

Ive been trying to grow mycelium in some sort of liquid substrate, so that i can make mycelium syringes but ive been having a few problems.
First off, i tried a suggestion on a post to use Karo light syrup.  I did as it said, but nothing ever showed up, even after a week in the incubator.  I heated the karo/water mix in the microwave to sterilize (the karo seemed to mix in with the water after heating and was not visible.  Maybe the dextrose molecule broke apart into something unusable by spores?)  I just shaved some of the spores from the print into the jar.  No contams showed up, but no mycelium did either.  Next i tried the same medium, but used a small chunk of colonized PF cake to see if the mycelium would grow out from it.  That solution got a little bit cloudy and smelled (contams?), but no strands of mycelium showed up.  Currently im trying another suggestion, honey.  I put about a tablespoon of honey in 3 jars, and sterilized them for 5, 15, and 40 minutes.  I did this to see if maybe the sugars were being broken into unusable parts by the heat.  I have yet to see the results of this experiment.  Any other suggestions as to why this isnt working for me?!?!?  Thanks alot!

I only ask, because I am not a fan of microwave sterilization (as - sometimes it does not).

This is what a healthy liquid culture looks like. Mix was 1/2 teaspoon light dry malt. 1/2 teaspoon dextrose to a quart of water, heated in a pot to a mild boil, then strained through coffee filters into a jar, then PC'ed around 15 pounds for 20 minutes.

Those are plated glass shards in the jar bottom (if you wonder). They are there to hel cut up the thick mass of myc - when the jar is agitated - so it does not plug a syringe).

Has anyone done it with just honey?  I see lots of recipes for malt and dextrose, but will just honey do the job?  I dont have ready access to those ingredients.  Thanks

Yes i have some Lipa Yai i did with just honey. I used the shittiest crystalized honey ever. I only sterilized it for 10 minutes... and i didn't incubate it lol. My culture sucks a dick compared to the one in the picture though. You want a 96% water to 4% sugar ratio.... 1tsp per 2 cups is what i used. It took about 3 days to see growth, but Lipa's are supposed to be fast colonizers.
