Cutting growth from agar;

Just a quick question:

How do you guys cut your healthy growth from agar for a transplant in tall half pint jars or quart jars?

I just attempted to do so with a scalpel but it was very messy and I'm pretty sure I compromised the jar to contaminants.

what was your issue exactly? you just have to cut a piece, and pry it from the remaining agar. you can also just shave the upper portion since the lower agar is not colonized. even sterilizing a hypodermic needle, cutting with it and transfering is feasible.

btw, having a flowhood when doing transfers is not necessary but helps a lot.

Well, I cut it out but couldn't get it up and out of the jar.  I ended up having to fool around with it for a while in order to press the growth on the side of the jar and drag it up.  All of this movement plus the amount of time I had it exposed to air (I don't have a flow hood and it was too hard to do inside of my glove box) makes me think it's going to get badly contaminated.

I have ordered some petri dishes in the meantime (and have some from a member here coming as a trade) but need a better solution for the growth that I have now.  I have about 8 half pint jars left to do.

I see, I didn't understand you were using tall jars as petri dishes for agar. if you can, use shallow jars. as of now best I can suggest is to either wait for petri dishes or use a needle to cut out, flip over and punch thru a healthy colonized piece.

also work with your jar sideways if you can, dust is always falling down and I think in this way you're a tiny tiny bit more clean than with the opening facing up. don't stir air up, try to work in your glovebox and use smaller jars or something better next time. :smile:

I might just trash the rest of these so as not to contaminate my entire work space and wait for the dishes to come in.
