Help Analyzing My Strain Isolates

This is my fist attempt at strain isolation. These are 1st transfers done 5 days ago and are the best looking 4 out of 25 dishes (they are all PE). I know I need to transfer further, but can any of these, such as D, be considered monoculture, ready to xfer to master slant, at this point?

those look nice....i think you want to test the cultures before you would make master slants..... save some of the labeled dish,grow it out and if it's what you want ,go back pull that culture to make slants.could be "A" or "E" or "D" that you want to store

D, possibly L and A. But to test you really need to grown them out on agar for a while and watch for sectoring carefully.
Me, I'd work with J, D, or F by taking small samples onto full plates and seeing what happens.

Ok thanks for the pointers - I'll get some full dishes growing and then grow some out from those.
@New Noon: These were from spore syringes that were innoc'd on malt extract agar in the dishes in that pic in my sig and xferred once from there.

thanks man working on agar myself same resipe and noc them up with ms syringes and they grew mys but it was realy transparent looking
just poured 20 more dishes like an hour ago goin to give it anouther shot in the morning

Yeah I've got a few like that too. The Amaz is looking good, but the GT and Ecu are all cottony/tomentose don't see any rhizo in them at all... not sure if they're worth transferring or not. I was hoping for some aggressive myc in those too but I may have to start from scratch for those strains.

i don't see very much sectoring on any of them dishes. did you use an lc from a vendor?

No they're multispore syringes from one of the most reputable vendors here though. They threw in an extra syringe of PF Classic, but that particular one petered out so I didn't transfer it.

You could fruit most of those now without further transferring. Often, the last few strains on a dish will merge during spawn run. I'd just use each section to inoculate a grain master, while transferring a tiny piece of each to new plates. Be sure to properly label all the plates and grain masters, so that once you determine the best strains, you can go back to the appropriate dish in the refrigerator and have that exact strain with the youngest possible cell lines. RR

Well, now you got "the word" and are good to go...
BTW, those cultures are effin' gorgeous. My PEU strain work so far hasn't turned up anything so rhyzo, but they grow and fruit really well (pic in my sig is the first flush from the first grow). Potency was well overboard as well.
Be sure to post a followup on how this work turns out!
