Popcorn contam? Possible causes?
    #15675525 - 01/16/12 08:53 PM (12 years, 5 months ago)

Hi all. First time tried using popcorn and unfortunately most of the jar's are showing contams. Wasn't too upset, I know popcorn isn't the best grain of choice, but ended up snagging a couple 2lb bags of it a few days ago until I can get rye berries. Just curious as to where I went wrong:

1)Made the jars using a lid with a 5/16" bit (closest thing to 3/8" I had) GE hole covered with easyfelt around the opening sealed with RTF as well as an additional RTF injection port.

2)Soaked for 24 hours

3)Rinsed then shook over the course of ~45-60mins to dry the outside.

4)Loaded into jars about 2/3 full, covered with two pieces of foil

5)PC'd for 60mins

6)Put sterile jars in glovebox, removed foil, wiped injection port, flamed syringe, wiped needle with 70%iso, injected ~2cc spore syringe

7)Put in TiT incubator @ ~78*F

A few days later is when I noticed the contams. Any ideas what went wrong?

I gotta try some pop corn. I bought some a long time ago with the intention and I'm starting to just eat it instead.

Anyway I would say your PC time is a little low, I like to do mine for 90 mins.

People are going to tell you the temp is a little high to probably which gave the contams a boost.

Edit: oh yeah if it was in just a few days it is prob the syringe.

Spore syringe from a sponsor from another forum, first time using them though... In case it was the syringe, I have a LC I can use for the next run. I have some more popcorn about to finish the soak tonight. I'll try PC'ing it for 90mins instead of 60. Also, when drying, any opinions as to just leave it in a strainer vs using a newspaper/towel?

I just leave it in a strainer, but staying on the dry side is better.

Toss it bro, kernels are busted and looks a little too wet, when i used popcorn back in the day mine didn't get busted and i would lay it out on a towel to dry ,dry to the touch but it was just soft enough where u could bite into it,seems like everyones turned away from popcorn now cause of contam issues. Anyway I thought people always pc's for 90 min with grain

Did you soak them in hot water?

I never got all those busted grains either but that was with soaking and simmering. Maybe the simmering helped soften them up some before PCing

Well I started try two with popcorn tonight. Did a long soak. Simmered for 60 mins again. Rinsed a lot more thoroughly than last time and dried longer. They're currently in the PC right now, on a 90min cycle. And as far as the jars, I put double the easy felt layer on instead of a single layer. I really need to get the whatman filters, from what I've read they work wonders. Any chance they're available locally or is it best to just get them online?

Also to figure out of those spore syringes were bad I'm just going to try it again on a couple jars and the rest with a pe6 LC.

Broken grains aren't an issue of you sterilize properly.  60 minutes will never cut it for sterilization.  People used to use cracked corn all the time and that shit turns to mush, and basically comes pre-burst.

On that note I should add: I used a different brand of popcorn this time. And glad I did 90 minutes this time as well

Popcorn just isn't as good for spawning because of the large grain size.

That's crazy looking! I just noc'ed up round two tonight, hopefully the longer PC cycle and the double filter layer helps. If not, then I'm switching to rye grain and getting some whatman filters. Anyone know of a cheap place to get the filters from?
