Prepping gallon jars, far too big for PC? For Mexicana sclerotia
    #15670277 - 01/15/12 03:38 PM (12 years, 5 months ago)

I have two gallon-ish jars with lids that I've got silicone injector ports and polyfil filters on. I plan to use these jars for a big batch of stones. I've got some RGS inoculated with multispore, and when they produce stones I'm going to clone a nice one to seed these large jars.

My question concerns preparing these huge jars. These jars are twice the size of my relatively small PC, so I could use some thoughts regarding how to go about this. I'll thoroughly clean and ISO the jars before loading the substrate, so how to prepare and inoculate the substrate is the question.

For the sake of its price & availability [and because the clone will have grown from it], I'll most assuredly be using RGS for these also.

Should I sterilize the RGS before loading and hope for the best, and inoculate with copious amounts of liquid culture?

Should I pasteurize the RGS, and if so, inoculate with either copious LC or lots of spawn [started from my clone] right before loading into the jars?

[Insert Shroomery's Option C]

Would it be a good idea, perhaps, to sterilize and load the RGS, then soak it with H2O2 and let that sit as much as a week for most of the peroxide to break down before inoculating LC? This could add one more step in favor of safety to neutralize any contaminants picked up while loading. I feel this may be pointless, so if you don't please say so.

None of the above.  Get a bigger pressure cooker or smaller jars.  You have to be sterile.

In addition, I've never had much luck with gallon jars.  They tend to go anaerobic in the center.  Furthermore, many strains only form stones around the edges, not in the center of the grains, so you're better off with filter patch bags that you can flatten out slightly for more surface area.

RR, thanks so much. That second piece of info changed it all.

Maybe sometime I'll try using these big suckers for fruiting Enoki!
