microwave + maltagar-powder + water + petridish = question

hi shroomers, hope you're all ready for new years eve.

i've got three glass-petridishes. is it possible to mix the agarmaltpowder with tap water (mix as in the package insert) — then placing the petridish in the microwave and heating until the mix cooks, stop and wait for cool down. put the plate into fruiting chamber (28°C)  (that all the resistant spores can fruit) after 24hours repeat the process to kill the fruited spores.


logical-thinking: it has to work. but whats your experience? thoughts?

Would the microwave be enough to sterilize the agar mix?

I don't think the microwave will give you consistent results with sterilizing. And as for waiting for endospores to germinate (I think that is what you mean by resistant spores) how will you know when the endospores have germinated and not made new endospores? I also don't think endospore are as much of a problem in agar mixes as they are on grains for example. You would have more consistent success using a pressure cooker. Also putting a plate in your fruiting chamber and waiting for it to contaminate I don't think is the best idea for future grows. Just my thoughts but feel free to experiment I suppose. generally, i use the pressurecooker, but i see following problem:

glass petridish: the lid sits just on the dish. when i cook the dishes with the maltagar/water-mix for 45min, hot air rises under the lid into the dishes. the mix cooks out of the dishes etc.

when i use the method of a "englerkolben" (cooking the maltagarmix first in this "englerkolben" then filling up the dishes... the dishes needs to be sterilized first, then they need to be opened in our glovebox, filled up.. etc. i think too many parts for contamn.

in the past three months we had a lot of contamns. i read about the "microwaves" they should kill through the waves microbiology pretty fast but it's not a common method. it's a method to making preserves.

i thought to ask here for experience of people who uses this method :-)
and if not: i'll experiment and share mine.
