So its been about 48 hours since ive inoculated my jars. Ive had my jars on a 12/12 light cycle to pick up rhythm and all that. I didn't plan on checking on them for like a week or so but eh I decided to ever so careful inspect one of my jars.
And voila. About a 2-3 mm spread of white fuzzy goodness. I dont really have any pictures, but I decided to start my post now seeing its only been 48 hours and im rather excited to actually see something take way.
Hopefully ill return to this thread in about a week with some nice looking pictures of some colonization. This is going to be my first grow, but I have put a lot of time and patience into it, and feel as though my procedure was immaculate for a first timer. Hopefully ill see some really nice flushes. I plan on consolidating the cakes until I see the first pin in any of them. My goal is to then print that very first pin I see. Thats my goal

your jars dont need light, I would put them in a box or some place with no light.

They don't need light but light is fine, you don't need to put them in the dark.

Thanks for the input. But ive done enough research to hear enough people say that ambient light helps. A lot of people say it really doesnt matter, and flushes can turn out on par with ones colonized in light and in dark. But I believe big on a thing about circadian rhythm and honestly in the natural, mushies are always on their 12/12 light cycle, or whatever actual light cycle, so Im gonna keep them on the 12/12 until I harvest.
Also why would I want to put them in a box. For them to colonize they still need GE, putting them in a box would leave stale air, granted no FAE is needed, but I have them in my closet where I plan to grow, it gets a little bit of cycled fresh air, I dont know why you debunk lighting so much.
Thanks for the opinions though.

Yea I know they dont need light. I just feel like natural rhythm is a big part of it.

I never thought of the whole circadian cycle thing, good thinking
and yeah I stand corrected light can go either way during colonization.

God upon further inspection there is fuzzies in almost every inoculation site. Out of 5 jars I see 3 "inoculation areas" that don't have fuzzies. So 17/20 are already showing signs of life after 48 hours im stoked. I really like the simplicity of inoculating on the glass so you can see it start out. Now time to see if the sterility paid off.

i smoke green bud too. (haha i figured i'd say that so you recognize who i am)
sounding great so far man. yeah, i'm with you on the ambient light thing. Why do people keep them in the dark the whole time when its not the wild they get a little (or sometimes a lot)?
As far as the box idea suggested: I wouldnt say put them in a sealed or closed lid box (then we're losing our light) but maybe a cardboard box with the lid loosed pressed down or a tub with the lid not snapped down. (I do the latter method and it works great.) The box is great for keeping the temps a little warmer and constant.

Haha hey man thanks for the help on chat btw, I think I left without thanking you. I think next grow im gonna try to colonize in the dark just to have photos and experience to share. Yea I have them set up in a small closet with the doors almost always close. Its been staying about 75-78f, I think next time ill try and colonize them setting them in a cardboard box with an open top. Just to see the outcome compared to not.
And ill try to get some pictures up soon. I dont have a camera anymore, but my roommates do so when they get back from Christmas stuff ill use theirs, I dont feel like using my laptops webcam, it is the shittiest quality.

Ok I decided to take a picture with my laptop. Its been about 60 hours since inoculation now, and heres a picture with the largest fuzzy spread I can find. Its about the size of an air soft BB maybe a little bigger. And you can tell how white it is against the color of the Verm and my gloves.

That`s how mine started too, leave em somewhere that isn`t totally dark all the time, I left my jars for 4 days, when I get back the growth will probably be surprising considering when I come back from work I am always shocked lol

So its been exactly 4 days, 23 hours since inoculation and jars are showing about 5% colonization at each injection site. Pretty consistent in each jar, 5 total. Each jar took all 4 inoculations very well, only one site isnt showing signs of growth, not really concerned. Ill have some pictures up soon once they have a little more time.

I'm gonna have to assume that Pins is saying your grow is contaminated because of the short time it's taken to show so much growth. Had you inoculated with LC, this rate of growth could be expected. Spores, however, can take up to a week just to fully germinate.
I've seen many people post on here claiming their jars reached 100% in 3 days. Well, yeah, perhaps they're jars did colonize that fast... just not with the mycelium they were hoping for lol.
I've been learning a lot lately about just how much a contam can look like the good stuff. I'm considering changing my technique to only birthing cakes or using spawn once I see knots/primordea/pins starting to form inside the jar. This seems like a safe way to ensure a good grow and clean work area.
Best of luck to you!

his point has merit... spores should most certianly NOT show growth after 2 days. did you flame your needle?

This is an albino MS after only 1 1/2 weeks:

I made the syringe from a print but the syringe was packed with spores so much so the water was dark.
