Tissue Culture / Microponics
    #1556852 - 05/17/03 10:45 PM (13 years, 6 months ago)

I've been reading about this as a means of propagating Cannabis. It seems like the seed banks should be selling encapsulated sterile stem pieces along with their seeds. It should be the absolute best way to distribute the best of the best genetics on a level that would be more practical than seeds.

http://www.overgrow.com/edge/showthread/t-166221.html

Most of the work would be done by the seed bank, what the buyer would recieve would be sterile, ready to root stem pieces in a small vial suspended in a medium. This would then be grown out in a specialized rooting chamber.

That way people could buy exactly what they want instead of growing out dozens of seeds, and picking through them to find the best mother. Anybody (that could afford it) would be able to immediately start growing that special one in a hundred plant.

Microponics looks a bit easier than tissue culture. It seems to be something closer to cloning only with a piece of stem without leaves.

Yep, its really just a tiny cutting. The main differecne being you can get 30+
clones from a cutting sized piece of stem and 100+ from tissue culture.
I have though a bit about tissue culture since getting a flow hood
and learning to play with agar.
Hard to do, as is microponic culture, but a bit of work with orchid seed flasks
should set you up to be able to work with microponic clones.
Easy to mail ext.Good idear :-) hope the sensi seed bank start playing about with them :-)

It would really rock if you could order specific phenotypes, like genius, or the rare hallucinogenic haze pheno.

This is a very interesting topic!  I saw a thread about this in the contam forum a while back, and I am now very interested.  A couple books were named in that thread.  Would anyone happen to have an extra copy to trade?  I was not aware that from this plant tissur culture, roots can be grown, and then an actual plant.  This is entirely amazing to me!!  Does anyone think/ know if this would work with cacti?

I'm going to start doing some research now!

so theoretically you can grow a plant via nute solution and a very small piece of plant? how do you propagate root from the culture?

That link contains all the information that I have found so far.

Right now I'm more interested in growing a plant from a small leafless stem segment an inch or two long, but using tissue culturing techniques it is possible to get a plant from just a small piece of leaf.

Teonan: I can't get Cannabis Culture around here. I don't even know of any place that sells High Times.

I may do some experiments in a few months on tomato cuttings. If so I will post the results.

Propagating roots from a culture is rather hard, and some don't take.

There are basically three ways to culture plants:

1) one for shoots
2) one fo a callous (fluffy white non descript parenchyma cells)
3) roots

two get them depends on the ratio of IAA to CK, IAA being rooting hormone and CK being shooting hormone. CK = Cytokinin and IAA = Indole acetic acid.

You'd want a shoot producing hormone mix, i believe it's [CK]>[IAA] you need both hormones though for the shoots to come, plain CK doesn't do anything. So you'd get shoots and then turn them into cuttings.

Plant Tissue Culture work isn't easy.
