checking sterility
    #1544508 - 05/13/03 07:52 AM (20 years, 9 months ago)

My friend's helper monkey was thinking of making some prints and eventually some spore syringes. He was concerned about sterility of the prints/syringes (after all, he is a monkey and likes to throw his shit at people). Short of innoculating a test jar with the newly made syringes or prints, is there a good way to check their sterility?

You can go to agar or knock up a LC with it, as a test run. 6T

Germinate on water agar and transfer periphery to nutrient agar. More info if you need.... I make bad spore prints. I'm lazy.

Alright, thanks for the info.
So in your experience, how often do the prints or syringes you make turn out to be contamed? From what I've read about print/syringe making it seems like it would be pretty easy for contams to get in.

It is easy for contamination to get in; I'm sure if I tried to make syringes out of my prints, half my stuff would contam. It depends on how you take it, though; a print taken from a cake from a clean enviornment not too long after the veil has broken should be relatively clean; I have even heard of people putting it on one dish, letting it sporulate a bit and then transferring to a clean dish so only fresh spores are obtained. I have also heard of people washing the caps in iodine and handling them with sterilized toothpicks.

I get dirty prints from casings, though, and then just make water agar (20g non-nutrified agar + 1L Distilled water). I then (under a flowhood) streak the water agar with a visible amount of spores (has to be a good amount.) These then germinate and grow out a bit, but anything else that is on the dish will not really grow. You can then take some mycelium from the outer perimiter of the growth (not where you streaked) and transfer it to a nutritious agar to let it grow (under a flowhood.) I don't have any problems with contamination doing it this way. I just recently cultured some mycelium off water agar plates that were a month or two old (I forgot about them and left them at R.T.) and it still grew!

i havent had any problems makeing prints and syringes and i'm not that carefull aboout it, to make a print i just put a shroom cap on top of some card with tin foil around it (i dont sterilize it i just use the in side side of the tin foil) and put a glass over it and when i make a syringe i dont use a flow hood i just clean the side and flame the needle, syringe and blade
