The problem is that people see BS and follow it. Even if it's been tried and failed over and over again.

The old stuff isn't erased forever. It was archived away from naive eyes.

This has been said several times throughout the thread. I guess you guys didn't read it or skimmed over it?

If you'd like include things from the "community>links>vendors" drop down menu on the site header..

There are a lot of vendors that are no longer running.

Yes and yes.

If I were buying my 1/2 pint jars again, I would probably lean toward
the wide-mouth variety, but this is splitting hairs.

Good luck,

To inoculate with a spore syringe (multispore) simply flame needle, wipe with alcohol,  and plunge syringe in through the polyfill,  tyvek and inject 1 (one) drop of spore solution in the center of media.
Remove syringe, replace foil and incubate at 80°F - 85°F.

This is a great, I love the method, but there is some incorrect info.

You may get away with wiping after flaming, there were times I did myself, but it's not proper.

Though when I was really concerned about contams I would wipe, flame, inject, wipe, then cap using a lysol wipe. That may have been overkill but it certainly wasn't risky imo.  But of course before injection, nothing but a torch touches the needle.

Also incubating, especially at 80F+ is also outdated.

Of course, silly me. That didn't really jump out at me the way the other two parts did. Didn't even notice it. Though I never put my foil back on, I just toss it in the recycle.

Ya, Well we can't all be pimps all at once. :pimp: I want those BAD.

I've done this off and on for years. I can't believe how much things have changed though. This time I don't plan on fucking off and falling out of the loop of the OMC.

Over the years I've used tyvek(both the right grades, and the wrong ones, house wrap  opps), polyfil, and micropore. Believe me, I'd like to get SFD's, but only recently did I right my ship finances wise.

I have a whole list of myco supplies actually, and it's ever expanding. Simple things I can get around town for cheap, like a 60ml syringe at the feed store.

All the way to a flowhood. I'd build a flowhood if I could afford it atm,  But in the next couple of months, I just want to make some realistic purchases, and trust me SFD's are on the top of that list, along with a proper inoculation loop, myco scapel(I think maybe I should spring for a long "fancy" one instead of just the plant store scalpels) I'm just now starting to agar. Cooking my first plates tonight actually, well technically today, cause I am the night. :batman:

Re: growing/page in General Cultivation, subtopic Sterilization and Pasteurization [Re: Lab Cat]
    #17293408 - 11/27/12 08:50 PM (11 years, 3 months ago)

There is a pictorial in sterilization and pasteurization showing how to pasteurize with oven bags. I am confused about one of the methods shown,as it says to pc the dung and straw for 90 minutes. I need to know if this will work or is that sterilizing the substrate? If that's the case can I pasteurize what I have left and mix the sterilized substrate with it to make an adequate amount? Any help would be sincerely appreciated! I have a jar of colonized wbs that is trying to fruit in the jar and need to spawn asap.

Your post confused me...what species are you working with?

If you can spawn to a filter patch sack (do you have a flowhood?) then
a sterilized sub will work fine.

Pasteurized subs are mandatory for some species, as they require "live" substrates.

Really there are many routes here.

Please post a link to the thread that give you advice and confusion.

We can help.

Take care,

JD

This was my first such edit, but I think that it looks good.

Thanks for letting us know.

I didn't see anything about cloning, or isolating srains.

There is a cloning section, but it is empty.

There isnt alot of bad information in this, but some of it is misleading, mainly about if theres moisture on the walls then humidity is too high for cased cakes (it may say casings rather than cased grain)

Im actually about to use this tek, so its not all bad. just using it as a refresher for doing straight rice which ive done before.

not to sure about this tek, plus there are some misnformed comments to the tek stating that shotgun fruiting chambers dont work in less than 50% ambient humidity and some bullshit about drilling holes in your chamber to be bad for humidity and temperature.
