PokeSmot - thanks for reminding me, your SporeDram tek is very similar to something I have done in the past and that works very well, and doesn't require the need for any other equipment besides what you would already have laying around.

Take one of your 1/2 pint jars and a spore print (see note about jars/prints towards bottom of post). Make sure the jar is sterile, and fill it with boiled/sterilized water. Cap it and wait for it to cool down.

Now for the simple part that no one seems to have yet stumbled across - take your print and wipe around the edges with an alcahol dampened paper towel. Next, lay it (print side down) over the top of the jar, and screw the cap back on (quickly, so as not to allow any contaminants to enter your jar). Shake the jar vigerously, and there you go - a nice, large amount of spore solution that can then be diluted as-needed depending on the size/darkness of the print used. Simply stick your syringe needle thru the injection site, withdraw some fluid, and dilute if necessary. (Obviously the jar preparation should be done in sterile conditions, otherwise you are surely going to get a contaminant in your solution.)

NOTE ON PRINTS: This will work with a print on foil OR on wax paper/other media, but you must follow the instructions on jar preparation below. Also, the size of the media the print was made on must be large enough to fit over the mouth of the jar, so that you can screw the lid on over it. I'm sure you could just drop the print in the water and not have any problems, but I have found that the backside of the media that the print was made on is more prone to contaminates. By placing the print face down over the mouth of the jar and screwing the lid on, you are reducing the odds of a contaminant on the other side of the foil/paper getting into your solution.

NOTE ON JARS: The jar(s) you use must have plastic lids or one-piece metal lids to keep water from escaping/contaminants from entering. A drop of caulk on the injection site makes for a nice self-healing injection port, and eliminates any chance of water leakage or foriegn matter entering your solution. Again, I must stress that it is much better to use a print with a large enough piece of foil/paper to fit over the mouth of the jar, so that you aren't exposing the solution to the outsides of the foil that you have handled and that have been exposed to outside air.

My foaf just makes invitro spore solution by hanging a cap in this gizmo - aseptic conditions & with F/hood.

Three dollar 1/2 gallon sun tea jar w/spigit. Spigot removed & replaced with resealing injection port. Lid retro-fitted with double tyvek filters & clip to hang cap from. Whole giizmo will PC with water inside (plus wetting agent).

Works well in a fridge. Cap does not rot & will drop spores for days. You learn quickly, when removing cap, to twist jar & hold lid steady.

Works well, 6T  :wink:

Sounds like that would work just as well - printing directly into the jar would eliminate any "in-between" steps and thus reduce your odds of contamination once more.

However, the print will tend to stick to the bottom of the jar (same for foil). The way I have found to reduce the "stickiness" of the print is to try and keep it as dry as possible during printing and after. If you don't fill the jar up with water to the top, you have plenty of room to shake the hell out of it, and most of the spores will come loose. Whatever doesn't really doesn't matter, after all, this should yeild plenty of spore solution until your next harvest. :smile:

The hippie uses 1/4 pint wide mouth ball jars with a plastic lid(one hole). Using 1/4 pint jars the syringe reaches the bottom, He uses the head of the syrine to scrape the spores into the water, this gets rid of the coathanger!!

Water with jet dry is placed in the jug & the whole thing is PC'ed. After cooling to room temp, a mushroom cap is then hung in the jug under aseptic conditions. The spores drop into the water in the jug. They do not drop onto dry glass. 6T

appearantly it helps spores from not clumping or sticking to things. But I can not confirm this

it is a non-ionic, inert wetting agent. gret stuff. keeps spores from clumping, actually, keeps them suspended. it does a lot more tho. any tough spill/stain is far more easily cleaned with the use of it. try putting some in your toothbrush next time you brush your teeth!! j/k... i think.

Could you tell me 6T how you "suspend" the cap over the water??

I usually add a sterilized glass marble, or dime to get mine off the bottom of my jars.

It is a very clever idea SixTango!

what a friend does is:
When emptying a jar of substrate (do it carefully with the open end of the jar facing down at all times), then squirt a sterile syringe full of water into it, suck it up and you have a quick mycelium syringe.

You do not need a coathanger or other scraping device. I don't know who originally came up with the idea but it is possible to inject sterile water on top of the print in the jar and then freeze the water.

Thaw it halfway and swirl or even shake. The ice fragments will scrape the spores from the bottom.

I mean empty the jar's colonized substrate with the top facing down, then turn it rightside up in a cleaned fishtank on its side, squirt the syringe in and suck it up.  You knew what i meant tho.. :wink:

Yup. But once the water is frozen and then partly thawed the spores can be scraped from the bottom by shaking the jar. The ice shavings do it!
