Wbs questions
    #15193290 - 10/07/11 08:02 PM (13 years, 3 months ago)

Over the past 4 days I inoculated 17qt jars of wbs. This was my first time working w/ bulk spawn and I screwed up on a few things.
My pc only holds 7 at a time so I did 3 batches (came out to 17 cause I sterilized polyfill and some syringes once). I made self healing lids w/ silicon ports and squares of tyvek taped down w/ micropore tape for GE. I soaked the wbs for 24 hrs and simmered for 30 min. Then I let it drain for about 10 min, filled jars 3/4 full and PC'd for 90min. After the pc was cool enough I shook them up and put them back in the pc.
Next morning inoculated w/ 2.5cc spore solution each, in a bleached and ousted down room  in a sterilite box flipped sideways because I couldn't figure out a way to flame sterilize in a GB...
Needle was flamed and cooled w/ an alcohol wipe, but I had to take it out for a split second to get it into the port (kept missing threw the alc wipe). I had a face mask and long dish washing gloves on.
1. Do you think I soaked for too long? I read that endospores make more of themselves if soaked for too long. The temps were 65-75 past few days and it was always overnight.
2. I know I messed up on moisture content by not draining the wbs well, but only some of the jars look wet. Mostly the grains look too dry to me, but some jars  are so wet that they stick to the sides of the jars.
3. I still have more jars to knock up so except draining the grain for 30min, and holding the tyvek down w/ silicone instead of micro pore tape, what do you think I can improve?

Hey Dimo,

I think you should switch the tyvek for SFD (synthetic filter discs).

Spend a little time tracking down an inexpensive source for rye berries.  Much easier to work with.

Sound like you're off to a very good start.

Good luck to you.

Thanks!
Yeah I originally wanted to use rye but couldn't find any.
Is the only way to get SFDs buying them online?..im already ordering a lot of questionable :poop:, so it seems like a bad idea for now (im not so worried about contam as water content)

Btw, forgot to mention they are incubating in a TiT w/ a 5gal heater set to 79°.

Umm... They sound good. The bird seed should stick a lil'. If your simmering, you shouldn't really soak to much past 14 hours... Other wise you need to probably reduce the simmer time to prevent burst seeds. As long as there isn't any pooling and their aren't to many burst seeds every thing should work out fine.

Most people soak for 24 to 36 hours if they intend to skip the simmer. Though make sure rinse really well though. Give them a little time and they should be cool.

Grated, I never used rye berries-- BUT, WBS is $14 for 40lb and that's just down the street. Soak it for 12 to 14 hours (over night), rinse tell clean, bring bit pot to boil, move to cold burner while dumping in soaked and rinsed seed, wait 30min to an hour, rinse well and drain for two hours (for a normal sized colander/strainer), jar up and pc. I fail to see how it gets much easier then that...

But if you got a convenient and cheap source for rye berries... Why would use WBS?

Oh yeah, to help prevent sticking add some gypsum to the soak water... And simmer water.

K gotcha, is garden gypsum good? Its BB sized balls and supposedly saturated.

Gypsum helps with clumping. If the WBS is wet it will stick to the sides no matter what.

Garden gypsum

If it looks "too dry" then is perfect IME. The soak is where the grains absorb most of the water. I soak for 24 hrs. The simmer is something RR came up with to quick dry the grains by steam drying. It's not necessary but the grains do absorb slightly more moisture during the simmer (which is really just bringing the water to where it begins to boil and then removing from the heat and dumping it in a cloander/strainer) and it definitely cuts down on drying time. Too dry is always better than too wet when it comes to grains or bulk substrate, so always try to err on the side of too dry .... if you err at all.

I've had jars with moisture in them a bunch of time. As long as you don't see pooling on the bottom of the jar you are probably OK.

Most WBS teks are the same or very similar. Check out my sig.

I would use EZ Felt for your GE filter. That's what I use and quite a few people will vouch for it. It's very cheap and works well in a bunch of different ways.

thanks a lot braa :mushroom2::mushroom2::mushroom2::mushroom2::mushroom2:

when you had jars w/ noticable moisture in them, did you notice delayed germination(multispore)? Slower colonization?

No. The only time I see slow germination or colonization is when the water pools at the bottom.
