my first inoculation

This is just a report on how my first inoculation went with a few minor questions mixed in. I would very much appreciate some feedback if you notice anything I may have done wrong or some advice for my next time (tomorrow). Sorry if it's pretty long, hard for me to briefly describe something.

I finally was able to start my first experiment today with a bag of grain and spore syringe. While I was knocking up the bag with the spores I was shaking pretty bad from a combination of excitement and nervousness. I used a clear plastic container for my still air box/glovebox that I had upside down with the end sticking off the table a bit for me to reach through. I used some clorox disinfecting wipes that I used to wipe down the table and the box then sprayed some disinfecting spray on the table and in the box and around the room. After that I went and pulled up my sleeves and washed my hands and arms pretty good. I went back to the room with my supplies that I had set out on top of the box and the ground next to me including a lighter and a gauze pad with 70% isoproryl alcohol (I have heard 71% but 70% was all the store had, is that still ok?) which I soaked the gauze pad with and set in the box. I had no gloves but used the alcohol to rub on my hands. I got the lighter and syringe and took of the cap covering the needle. Once I had the lighter on I began heating the needle until the flame got really low because it was running out of fluid. It was my only lighter and it was to late to pause then to go back to the store, so, praying it wouldn't go out on me I begin to see steam coming out of the tip and it started to glow right as the lighter ran out. I moved the syringe in the box and squirted a bit out until it stopped steaming as it was coming out. While I did that with one hand I grabbed the gauze pad with the other and pressed it against the injection site (should I have rubbed it and not just pressed it there?). Once the needle had cooled down I placed it in the bag and squirted some of the spore solution inside. When I extracted the needle from the bag I replaced the alcohol pad on injection site while grabbing a piece of tape to place over it. I kneaded the bag before all of this to loosen it up a bit and also after inoculation but not as much, just very gently. After all was done I looked at the syringe and noticed I used up about 2cc's including what was used to cool down the needle (was that not enough? It is a 1lb bag, I do not know its volume).

Notes I will remember for next time and for other inexperienced people like myself:

    -Remain calm. Like I mentioned, I was kind of shaky which made it hard to control my tools and easy to touch a sterilized object against something.
    -Go over everything before beginning the process then go over it again. I kind of stalled between some steps. For example, after injecting the spore solution I did not have the gauze pad ready to replace until I got the tape so there was a few seconds between removing needle and covering the injection site which left a window for contams.
    -Make sure to have all supplies ready beforehand. After my lighter went out I realized I didn't even check to make sure it worked first. If there is something needed missing it will be better to wait a day to get it rather then risk proceeding without it.


Even if I get a contamination on my first attempt I do not believe I failed. I still gained something very valuable; Experience and mistakes which I have already learned from.

Overly cautious is a great way to start.  You should be good, hope you ordered from a good vendor.

and yes I did order from some great vendors. out grow, mad season, and freespores (and another non sponsor spore vendor that was kind of sketchy). but all the sponsers products are great.

Here is an update:
did 3 more inoculations today, was pretty fun but on the first two I did there were some small mistakes I made but hopefully nothing that will cause disaster. On the last one though I feel I did everything pretty good, except maybe not enough injected. But it's better to use to little then to much right?

I'll be making another report for my log once I start to see any changes, whether it is mycelium growth or a contam.
