Say somebody inject 4 ml of glc in a quart jar, then used this quart jar to G2G. would the mycelium be too weak? discuss.

assuming the GLC was made from a Jar you inoculated with a spores, you will be fine. I do this currently.
MS spore to small pint jar with WBS. Then GLC from the pint jar to Quart Jars.
Quart Jars to Bulk. Finally to fruiting.

To maximize your MS syringe...
1. Inoculate MS syringe (one cc) to one pint grain jar 3/4 full and let fully colonize
2. Extract GLC and inoculate into jar of ELME LC jar and set aside to expand
3. meanwhile; G2G pint jar to six 3/4 full quart jars
4. Once quart jars are colonized; G2G each quart jar to twelve more
5. Then spawn X jars to bulk substrate of choice
-------------------- Please assume any and all prints exchanged are "WILD" in nature; and thus, should NOT be considered ASEPTIC...

How well do you think this method would work with an isolated strain??

Even better!
Instead of step one being random MS, it would be agar wedge to pint jar to start off with... Some start their LCs with an agar isolate... Just make damn sure the wedge was clean and it's always best to test any LC with a couple grain jars or cakes, before inoculating ever jar in the house with it.

Is it possible to inoculate materials that was not pressure cooked in order to test jars? For example boiling the grains?

It is possible but not the best practice. The idea of testing a LC is to make sure the LC is clean of any contamination.
If you inoculate a test jar that you did not pressure cook and it becomes contaminated you will not know if is from the LC or the grain.
