1st timer cultivation - pre incubation Q
    #15169686 - 10/02/11 11:37 PM (13 years, 3 months ago)

So im a total noob at this and I cant seem to find my answers in archives.

Status:
I have inoculated my jars in open-air with flame and alcohol sterilization into 1/2 pint jars.

Questions:
From what I have read, everyone says to put the jars into a dark place. Is a sealed container a bad place to store them? At this stage do they need oxygen or light? My point with this is that I plan on storing them in the attic or something and I want to do my best to avoid contamination. So can i seal em up?

Also during this 20 days or however long its supposed to take, what kind of humidity should the environment be? I know the temp is supposed to be around 75-80 deg. F, but does humidity matter at this point?

Although, if the specifications I seek are documented for this particular stage in the process please point me there. Thanks!

open air huh

dont seal it up, you need GE (gas exchange)

do you have a dry verm barrier in your jars?

Just make sure the jars are off the ground, keep them at least 2 feet above the ground, where most of the contams hang out.  Cubes benefit from light (pref 5000-6500k temp; "daylight"), so just get into the rythem of 12 on 12 off, or get you a fancy timer thingie.

Colonization only need gas exchange (GE).. If used pf tek (4 noc holes, etc) and you used aluminum foil, when you noc them up, replace the foil w/ micro pore tape and leave the lid secure.  That will give you the GE you need w/o needing to worry about much/any free air exchange (FAE).  If you don't have any micro pore (NOT TRANS PORE, etc).. leave the foil on, loosen the jar lid a smidgen for GE, but not so much you're providing FAE.

A dry verm layer, if used, is typically good enough to keep contams from reaching your cake during colonization.. but if you're using the 4 hole pf tek (as I did), I strongly urge you to chuck the foil (after noc'ing) and just tape them up for GE.  I also open air noc'd, the only jar I lost to contams was the one who's lid popped off.  That's when I learned the difference between FAE, GE.. and what typical contams looks like, LOL

Pics, upload them to your shroomery space: http://www.shroomery.org/forums/upload.php?action=viewthumb

i wouldn't open the lid at all, and you don't need tape if you have a dry verm layer

Humidity is not important at this stage

Ideal colonization temp is 69-75

Tape is an extra measure, more than likely ok with out it as long as you have a dry verm layer where you wiped the inside lip before adding..holes out in open with no foil on top

4 inoculation holes in half pint wide mouths sized 1/16"-1/8" is sufficient for ge

Proof on not keeping them in the dark...lets grow vids

OK I will leave them as is then. I did do a 1/2 inch dry verm layer with the 4 holes, im afraid i f*cked the whole thing up though because i had to punch the holes bigger after pressure cooking to get the syringe in. I didnt realize it was so thick! But i did my best to sterilize EVERYTHING (e.g. the nail, the hammer, my gloves, etc.) I wore long sleeves, a hat, and a mask in a non-drafty zone.

Currently they are sitting in a closed tub on the highest shelf in my attic. I will see if I can get a light timer and one of those bulbs from a grow store later today.

Thanks for the input

I definitely would have sterilized those jars again after making the holes bigger. Hopefully everything goes well though!
