What to do with a petri dish bursting with life?

I have two petri dishes that I knocked up with some GT spores back in July. The plates are fully colonized to say the least. Surprisingly there are no green or discolored spots, just an even brown across both plates. I'm aware of the need for sterilisation etc, no need to stress that here. My question is what to do with what's in my dishes to grow them to the next level. Liquid culture? Grain LC?
I'm just wondering how to proceed here. Any help would be greatly appreciated.

personally, i would use a section to noc up a grain jar (or 6), then make grain lc from that.
you could just drop a piece into some lc, but im weird about opening my lc jars after they come out of the PC.
you actually have a TON of options here, but i would never suggest something that i dont have experience with.

Why is it brown? Have you isolated at all? That should be your first step if the brown isn't contamination, then wedge->grain master ->glc or g2g up to you.

Can you point me to refs on preparing a grain jar for impregnation? The method you had in mind.

Why is it brown; Dunno, I figured that was just what color it is. I've never grown in a petri dish before.
The brown doesn't look like contamination. It is very clear brown. Not turbid at all.

I, personally, would just do another transfer and look how it grows. Healthy cube myc is white, not brown.
EDIT: If the culture is this old and does not look healthy anymore (brown), you might just want to restart with a healthy young culture from spores. Is it an isolate? If so, try to save it by doing another transfer. MS tissue just isn't worth the work.

I would go GLC route as well, as long as your agar isnt contaminated
here's the best tek I know of...
much easier than G2G imo.

i would go light malt and yeast lc ..... some of my dishes look abit brown on the underside if i have left the petris for a long time

top of agar - alt ms (no contams)

bottom of dishes 1x alt7 ms and the other is lions mane - all my cubes are in storage in a culture fridge ... my lab is busy being cleaned so i dont want to go pull them out, when i do ill take pictures.

i think this is the brown he may be talking about.

Brown always seems to end up being a bacterial contamination to me but as said. Pictures halp a lot.

Yep, this is it.

I don't have the dishes with me or I'd take a pic. I actually gave them to someone who has never grown and I'm asking on their behalf. Next time I grow it'll be a few syringes of penis envy nice and fresh from my neighborhood spore shop.

I wouldn't touch those dishes with a thirty-nine-and-a-half foot pole if that is how they look from the tops!

At the very least just for experiments sake take a grain sized piece from a leading edge and transfer it to a fresh agar plate and see what happens
I definately wouldn't be doing any transfers to grain with brown mycelium though

Ok this is my opinion on what this brown is...... because in true fact it is not brown but i slightly darker colour of the MEA due to two reasons. 1st where the mycelium is thickest it is a darker colour, so less light can pass though causing a slightly darker spot.
2nd metabolism – common name – mycelium piss ...... have you ever noticed this on your cakes, trays or tubs ? same this happens on a petri. I normally notice this darker area before i see a pin form on a plate.
Please do correct me if the above is incorrect

I got bunches, but they arent sterile. They are just wrapped up in foil and have been all over the country with me, even while I was in the hospital for over a month.

If your spores were sterile I'd feel bad for you.  You could continue your agar experience and try to clean the culture obtained from the spores.  But I guess getting another print/syringe would be better in case you're lazy.

I'm not lazy, so much as I am busy. At the moment I just don't have the attention to spare something as delicate and precise and that would need to be. I'll be moving in a few weeks (again) and maybe then I'll feel up to something like that then, but now, no fucking way.
