Made a bonehead mistake with LC jars
    #15100309 - 09/19/11 12:52 PM (10 years, 1 month ago)

Last week I PC'd some jars of honey solution to make LC. Normally I would drill my inoc. holes but I used a little nail. Unfortunately, when it came time to inoculate the jars, I realized the holes were a bit too small to get my syringe tip fully into, although I was able to get enough in to be able to inject. Now my LC is almost fully colonized and it's just finally occurring to my dumb ass that I'm going to have an even bigger problem in reverse.

So, should I try to get smaller gauge needles? Risk contams by slightly opening the jar to get at the solution? Start over from scratch?

If you can get a smaller gauge needle that won't be a pain to use with a LC, then i'd do that. If not then open out the holes a bit while being as sterile as possible. You'll need to test the LC either way on one jar to see if it's clean, so go with what's convenient for you

good luck!

I think once the mycelium has colonized fully, it's not as susceptible to contamination. You should be ok opening the jar if need be, I would still try to be as sterile as possible, but it should be ok.

That is true of grains & cakes, but not LCs they are extremely easy to contaminate.

I would do this, if you have tinfoil over the lids you can rub on it to see where the hole is. I would get a new nail and hold it in a pliers and get it red hot, then open up the hole a bit more, pushing right through the tinfoil. Then I would put on new tinfoil or rotate the original foil around. Do it in as clean a place as possible.

Bad idea, your mycelium may be fine with contams in the LC, but you still have to shoot it into a jar where contams will ruin your jars.

That's true for jars of grain, cakes and bulk substrates, but if you open a jar of LC, even if it is fully colonised it will likely contaminate, unless this is done in a sterile enviroment.

This sounds like a pretty good plan.

Thanks for the tips, everyone. I'll be giving the nail trick a shot.
