Cobweb prevention?
    #14945160 - 08/18/11 11:49 PM (12 years, 7 months ago)

So i posted this in contam forum with not much activity. I have searched but couldn't find much on prevention.

So just check my jars, had 10 lost 7 all to cobweb.
What precautions can i take to fight off getting a  cobweb contam? so far i have lost all jars except 3 to damned cobweb.
got a fresh batch sterilizing as we speak.

Make a glove box and develop a better sterile technique?

hi Nox overview your sterile technique. i used to have bad problems with contam and thought i was doing everything right. i sat down and assessed my sterile technique and noticed i had picked up one or two overlooked bad habits. i then went back to basics to relearn techniques by the book and slowly adding my own flair again.
my success rate is now much better. i get about 2% contam rate on grows. 100% success on agar,grain,g2g,lc's etc...

hope this helps
peace

Well i redone my 7 jars today.

used a different room to innoc this time. Cleaned it all down and its alot smaller.
used different source of spores for each innoc and left 2 just sterile to see if its my substrate/sterilization process.

I use mine upside down... Meaning; the bottom of the lid is used as my work surface and the bin is placed over what I'm working on...

Some have done so with the bin on its side and placed their arm holes through the lid.

I would be suspect of your syringe, along with reviewing your aseptic tek.

You are over thinking the GB.

Get a 105qt tub, and a 6" hole saw, and your done.

Just use disposable latex gloves, and clean the box before use.
Wipe down each jar, and tool as you put it in.

Its not the syringes, freshie every jar. I make my own maybe thats my problem in that process. They are not prints, but the mushroom caps themselves are my source.

So a GB doesn't have to be airtight?

Ahh k, will go buy myself a tote.

Quote:

Wild samples, alot of different sources.
150ml cups. Verm, Manure and BRF with foil folded twice to achieve 4 layers. This is done twice so there is 2 foil lids that are 4 layers thick.
Half inch of dry verm as barrier now been increased to 1 inch.

Boiled/Steamed in pot for at least 2 hours.
everything gets a alcho wipedown, hands washed with antibacterial soap.
bleach and glen twenty sprayed in air an left for 5 mins.

Innoced in my kitchen after a thorough cleaning with bleach, now in a pantry of sorts that got the same treatment with twice the bleach content.

No glove box yet or PC because i went to purchase one that had been there for ages at big discount but was sold day before and only one they had was with no gauge.

Gonna hit OP shops and 2nd hand electrical an furniture.

Scape the gills with a knife that was boiled/bleached/alch wipe an flame off, into a spoon that had same treatment as knife.
Sterile water that was done while boiling jars.
Open syringe suck up solution minus big 'bits'
Remove top lid
wipe needle with alch and injection site. done for every injection.
replace top lid

This sounds like where you may be having a problem.

Why not just print the caps under a glass?

I PC my syringes in qt jars.

Swabs are best done to agar, as it's easier to spot contamination.

These are BRF cakes..... correct??

And you are following the tek perfectly?

Dry verm layer and all?

Where are you getting these caps?  In door cake......... or wild?
