Hello... I'm sure I have read this somewhere but now that I need the information I can't find it anywhere. I'm pouring agar plates, but the past few times I've found that the agar quickly finds itself on the inside of the PC.

I fill pint jars (thats all that fits in this PC till the new one arrives) with the mix, about 125ml's of solution (which is less than half full). Then I cover with tinfoil and PC for 45 minutes. But even so, I keep finding the agar bubbling out of the containers and all that I am left with is less than 100ml's of agar, and gooey shit coating the PC :frown:

Any ideas?

is it really nescissairy to sterilize agar in a pc? can't you just put it in your microwave? that way you can easier control the 'overflowing' of the agar...

I used to use Quart jars with a tyvek filter disc and the lid almost tight.
Worked like a charm.

I'm sure if you did the same with a Pint jar you would be fine.

yes, you can sterilize agar in a microwave .
do small batches, and add a little extra water .
nuke 3-4 min. at a time, and watch out for boil over .
i have a dinky 700 watt m/w so i nuke a half full pint jar for a total of 8 min.

I would guess that your PC is cooling off too fast. That would cause vigorous boiling as the pressure drops, causing your boiling over problem.

road, it also works to use those juice bottles with graduated necks (for ease of pouring) then puncturing the lid and stuffing with pollyfill.

I do it exactly like Roadkill.
Tyvek filter is great also because it filters air as it enters the jar when it's cooling down.

It happens if the pressure cooker cools down too fast. Make sure to let it naturally cool down slowly until the pressure is back to atmospheric. Only then open the PC and take the bottle(jar) out.
This is usually a problem with thicker agar formulations, for instance oatmeal agar or dogfood agar. Less common with PDA or MEA.

Fifteen minutes of sterilizing in a PC is usually enough for agar.
Instead of pouring petri dishes, I prefer to use flat sided bottles...liqueur bottles or prescription bottles. Microwaves are great for preparing and melting agar but they do a poor job of sterilizing. I like to put about 10 or 15 ml of freshly prepared agar into flat sided bottles and sterilize for 15 min in a PC and then let the PC cool SLOWLY. When the PC has cooled enough to handle the contents, remove the bottles and lay them on their sides until the agar is nice and solid. The bottles can be stored for many months on a shelf. It is a bit of a problem to work with agar through the narrow neck of a bottle but it is not hard to fashion tools for that purpose from heavy wire or bicycle spokes.

Put Agar in flask (erlenmyer), stuff cotton-ball in neck of flask, swirl, put in microwave.

Nuke the bitches for 15 minutes at power 30.

Pour onto plates.

Another thing that will make your agar overflow is if the heat is up too high during sterilization. Anytime steam is escaping the top of your pc, it is also escaping your agar. Once you reach 15lbs, turn the heat down as low as possible to maintain that pressure. Since the pressure inside the agar bottle is also at 15psi, anytime the little weight on the top of the PC rattles and lets off steam, the agar is allowed to boil in the bottle. This causes it to 'boil over' just like a pot of sauce on the stovetop. And of course, like was said earlier, if the pc cools down to fast, the pressure in the agar bottle is higher than the pressure in the PC, and it boils over.

I use a screw cap elrenmeyer, tyvek under the cap and the cap screwed about half way down...I never worry about boilover....I got about 18 of those....250ml to 1000ml...they work well....

With that AA PC, if it has the little stopcock type of bleedoff valve (mine does) you merely need to unscrew it a little bit until it starts to bleed off steam at the end of your sterilisation cycle. It takes mine about 5 minutes to 7 minutes to bleed off from 15-17 lbs/". That slow rate will prevent boilover caused by rapid depressurization at cycle end.
