wet bubble- what to do?
    #1490564 - 04/24/03 07:49 PM (14 years, 6 months ago)

some time ago i got a print from a local strain of P cubensis , after isolating it in PDYA i inoculated a JAR of popcorn, all went well but i noted that the agar dish developed wet bubble.
So after cased (coir/verm 50/50), the popcorn developed it too and there is growing some strange fruitbody that looks like some mini white osters (i mean realy strange) so what should i do?:
Should i try a new multispore inoculation?
Should i transfer some of the culture to peroxidated agar?

and just one more qst:
What is wet bubble (fingi or bacteria)?
thanks.

I must be missing something... you claim to have wet bubble, but you don't know what wet bubble is?

Regardless...

If the contamination grows slower than the cubensis mycelium, and you can tell them apart, you should be able to isolate a non-contaminated sector of cubensis mycelium on agar.  You will want to transfer the non-contaminated sector to a new petri dish and grow it out to ensure that you did not transfer the contamination.

You say the contamination grows strange mini-white oyster-like fruitbodies, then it is probably a fungus.  You won't be able to use an antibiotic to kill it off.  Peroxide will probably not help much either.

If you can get the cubensis to fruit along with the contamination,  you could try to clone from the cubensis fruitbody to isolate non-contaminated cubensis mycelium.

If you have a good microscope, and good lab technique, you can make a super dilute spore solution and grow out only one or two individual monokaryotic mycelium sectors per dish.  Identify the non-contamination mycelium and transfer these to a second dish to form dikaryotic growth.  Its a lot of work, but it may be worth it if you are working with a new race.

Respondendo a:

I saw some pictures by the net about it and i m sure that is it! (amber bubbles, kind of ammonia smell, all simptoms...)

Respondendo a:

i can't, the dish looks like a pure offwhite mycelium when inoculated but when it gets older it starts to develop the disease.

Respondendo a:

i think this is the P cubensis fruitbody! the literature i've searched talk about deformed fruitbody from cultures of agaricus which developed this disease.

Respondendo a:

If you have a good microscope, and good lab technique...

well at least i'll try... thanks

i don't have a digicam so no pics...

Ah, I was assuming the odd shaped bodies were a contamination, not deformed cubensis. If your contamination is bacterial in nature, then agar with antibiotics added will help cure the problem. You can get premixed MDA with antibiotics added from fungifun (staments site). Grow out the mycelium on the agar, and transfer a clean wedge to the substrate for innoculation. Hydrogen peroxide might also help, but not as well. Good luck!
