Time to learn a new skill!

I'm finally ready to start the spores I took from some GT I had like 1.5-2 years ago. They are just wrapped up in foil and no doubt contaminated. I bought a huge bag of sterile petri dishes and ready to pour agar to get them started. Seems like I remember reading about using dirty spores a while back, and how on agar in a petri was the preferred method since it allowed you to then use a sterile exacto knife and cut out healthy desirable/fungal growth which could then be spawned on a larger scale. I'm not holding a ton of hope for them. If it works, great, I get to learn a new skill and I'll have what I already know to be some pretty active lil' knot heads. On the other hand, I'm already planning on ordering a few fresh syringes of APE soon anyway. So.....

The fewer spores per petri dish, the better... Well; at least two health ones, lol...


From what I've studied; petri work is THE way it's done :smile: You should be fine. This is how wild specimens are "cleaned up", too (spore and tissue, alike)  :smile:

You've obviously never seen the hilarious video of me taking those prints. It was more like, I'm going to eat these, but I should save some of spores for the hell of it. I was not trying to be all sterile and whatnot. So, these have been contaminated from the get go.

Yeah, you're right, I wouldn't worry about trying to keep that print clean!  :lmafo:

Yes, agar is definitely the way to go here.  But, I hope you've made a glove box for the agar work.  It's pretty unforgiving if your clean techniques aren't followed carefully.  The problem is that competitors like bacteria, trich, etc. grow faster than those spores will germinate.  Give your spores the best possible chance to get established without contams, or else it will be a long, tough battle to get a clean mycelium sample.

I'm good at making glove boxes lol. I have a natural talent for that, just not for taking clean prints.

Remember it can be a good few days before you see any activity from the spores.  I spread some spores on agar at least a week ago, and it was only yesterday I saw the first growth (but they're going fast now)

I often wish I had one.  A good microscope would add a new dimension to this hobby.

Unfortunately I spend too much dough building extravagantly expensive flow hoods and stuff like that.

I saw a pretty nice digital one with usb for $150 at the science store the other day. That's not sooo bad.

You want a minimum of 400 x magnification for Mycology :smile:

Are you serious about 400x or are just encouraging me to buy badass toys? I had a closer look at the one for $150 and it's only like 40x or some shit. Realistically, how strong a scope do I need for a hobby involving fungus?

Here's my plates this morning. It's hilarious how many spores I put on there. Oh well, now I know. I have no shame in doing things fucked up the first couple times lol.
