Well, pizotifen is a 5-HT2A antagonist.  This means it binds to the receptors that psychedelics work on, and blocks action on these receptors.

But there are several reasons she could still be able to trip.

1. If the pizotifen has a lower affinity for the receptors than the particular psychedelic, the psychedelic will just knock the pizotifen off the receptor.

2. Just like an agonist causes receptor tolerance via overstimulation of the receptors, an antagonist can cause a receptor sensitivity via understimulation.

This seems really significant BothHands.  Pizotifen must help prevent headaches because the 5-HT2A receptors are constantly clogged up when someones on this medication.  Yet if a bad headache does occur my wife will immediately take a dose of  mushrooms and this will stop the attack completely.

It looks like pizotifen and psilocin are doing the same job then? only psilocin might be a for more effective 5-HT2A antagonist than pizotifen.

Well just having an extra weapon to fight headaches with must be an antidepressant in itself.

Apologies for sidetracking from OP.

No, psilocin is a 5-HT2A agonist, while pizotifen is a 5-HT2A antagonist.  They have opposite effects.  An agonist turns a receptor on, and an antagonist prevents a receptor from being turned on.

Why two complete opposite receptor effects have the same effect on headaches, I have absolutely no idea.  It's interesting though.

depression sucks x.x

I guess if it's some chemical imbalance that is making you feel like shit, you should probably correct it but if it is something that you can reason/rationalize for yourself and come to an understanding why it's illogical to be depressed then I would take that route.

as for me,
anime is my anti-depressant lol

How can you not be happy with all the moe ^^

If you want to get rid of your depression, prozac at best will turn you into a zombie, disconnected from the world (and the withdrawal effects can be fatal. Better to come off sooner rather than later).

There is a wealth of information on the effects of SSRI's online. They are horrible, horrible drugs that do not help with depression, just mask the symptons. For some people, that's all they want.

Personally, I'd ditch the prozac, trip on the mushrooms, and then think about your life, and what it is that's making you depressed. If your environment doesn't change, then prozac won't cure you, it'll just make you stop caring.

Certainly do not mix SSRI's with drugs that fit into the serotonin receptor sites.

I realize this is an old thread, but information doesn't age, so...

SWIM suffers from severe clinical depression and currently takes a daily regimen of:

60mg Prozac
8mg Valium
25mg Seroquel

SWIM, without taking any break from this regimen, ingested just 2g of mushrooms and had a glorious journey. SWIM has experience with many drugs over many years (LSD and mescaline among them) and would put this recent experience at least among the top, if not at the very top.

Not only was the trip not diminished, SWIM also enjoyed a respite from depression completely for a few days, then a gradual diminishing of the effects, but still weeks later retains some of the relief due to the catharsis involved in the spiritual cleansing.

Anyway, thought this second-hand tale was worth adding to the knowledge database.

Good for you....glad things went well....sure would be nice if you could tone some of that daily regimen down. It is, however, very surprising to me that 2 grams had such a effect with all that shit in you.:sunny::peace:M
