I've been working on this fucker for way too long, and finally finished this last weekend. I thought I'd share some basics, some pictures, and can address more specific questions as they arise.
Scienceguy's Flow Hood, "Flows Good Man"
Filter: 18"x24" HEPA from FP Fan: Dayton 1TDT2
I built the box itself out of 1/2" MDF core maple, and maple hardwood pieces for the face frames and hinged frames, as well as internal bracing/sealing strips. It's designed so that the pre-filter is easily changed by swinging open the frame on the top box, and to change the HEPA filter one would only need to unscrew the top box and lift it away to be able to slide the filter up and out vertically. I have access to the fan right behind the pre-filter. Sash locks keep both top and bottom front frames securely fastened.
The whole thing was finished with Danish oil and then treated with a citrus carnauba wax. It's a living finish, constantly changing and deepening in character, and one that can be easily renewed. It's also hard as fuck and waterproof.
The frames were all pocket screwed and glued together, and the face frames were also pocket screwed onto the actual box. The box itself uses miter joints to increase strength, squareness, and the overall aesthetic, and because butt joints are whack. All joints are glued, many are glued and screwed. In some cases it was for added strength, in most cases it was to add stability so that I could still work with it before the glue really set up.
The plastic pieces are Lexan, cut on the miter saw and rounded out to fit the routed spaces for them inside the hinged frames.
I used an old work box and a gasketed, outdoor rated switch cover that does a great job of sealing in air. I exited the junction box on top of the fan with a 1/2' indoor strain relief, and exited the box itself with a 1/2" liquid tight strain relief. I used a heavy duty 15/20A switch more to match color than anything else, and it's total overkill. The whole thing is wired with a 16ft extension cord that I cut the female end off of.

BTW I was watching a Kreg joints infomercial the other day and thought this was something I MUST have.

Steal away! It's for the taking. The whole point is to continue to inspire good design.
I did actually use the Kreg jig for this project. I've seen other pocket hole jigs, and some look good too, but my buddy with the wood shop already had it, and it worked really well. Definitely a must for finish carpentry.
@NCDOC This is where the air filter came from. It was about 200 bucks shipped.
There are specs available for the blower here, though I ended up getting it from somewhere else for a little less than $150.
Furnace filter: $10 Hardware, electrical shit, hinges, etc: $50 Lexan: $40
I spent about $130-140 on the wood at a local hardwood supplier. They're really fantastic. I ended up getting all the hardwood pieces as 4 to 5 foot strips, instead of longer ones, for less than half per board-foot than they wanted for uncut pieces.
All in all, I'm sticking an investment cost of about $600 on it. If I'd have worked on it straight, I'd have had it finished in 3 or 4 days.
You could do it cheaper if you didn't build it out of maple, but I really consider maple the ideal choice for something like this without going too crazy or too cheap. I priced the same thing in cherry and the wood alone would have been almost $300, or you could build it out of OSB for less than $25. It came down to me just liking how maple looks, and how ungodly hard it is compared to most furniture woods when finished correctly.
@ others Thanks for the kudos! I really hope this just inspires some people to realize how easy and cheap even a furniture grade flowhood can be to build. The same size one from FP, as bare bones as it gets (but including a prefilter housing) still goes for $1100-$1200 shipped.

I shopped that thing together about three weeks before finally finishing the hood, but definitely well after I started. I didn't even have to make any letters...

Also, I neglected to mention the strip of electrical tape I ran down the inside of all the piano hinges on the lower box part, to keep dirty air from getting sucked into the sterile air stream via the venturi effect.
