Cultures contaminating in fridge.
    #14751666 - 07/11/11 02:19 PM (13 years, 6 months ago)

I have been wrapping my petris in glad cling wrap. They stay good when left at room temperature. After refrigerating (few weeks or longer) I get about a 70% contamination rate after I let them grow out a few days at room temp.

Do you think that parafilm can solve this issue?

Parafilm and placing each dish in a sterile baggie in front of your flow
hood after sealing.

Ziplocks and other baggies are sterile out of the bag, just wrap your
dish in parafilm, place into a freshly opened bag that was inflated from
your laminar flow and seal.

I NEVER get contamination from stored plates using the above mentioned
method.

Zip locking hundreds of petri dishes seems impractical, not to mention unnecessary.

I am willing to speculate that just parafilm will be sufficient.

You asked how not to get contamination, there is the method I use that
works %100 of the time.

Another reason for bagging plates is you can use less parafilm. If you
are leaving plates in open air you must wrap the plates 2-3 times around
to make sure that the film does not dry out and crack after weeks of
sitting and compromise your plate.

With bagging your plates you can get away with 1 wrap as the bag holds
in the moisture and stops it from drying out. I have used this method
for the last year and have yet to get a contaminated plate after storing.

RR also suggests this method. If you are using hundreds of plates then
just go and buy a few rolls of parafilm and wrap a few times around.

I purchase bags that are just big enough to fit a regular sized petri
in it for $1.45 at a local crafts store. They come in bags of 50 and
for a little bit extra work, and a few dollars I can assure my petris
are stored in a sterile baggie. Very practical, and totally necessary
as far as my lab techniques go.
