I keep filtering the LC and sediment keeps showing up!
    #14738941 - 07/08/11 11:39 PM (12 years, 7 months ago)

I'm not sure what the best was is to filter a liquid culture.  I boiled the water, mixed in the malt extract and dextrose, let it cool, and filtered it.  The filter didn't look like it took out too much.  Then I pressure cooked it and bottled it and put it aside, and the next day it was full of sediment.  Is this some sort of slow forming sediment, or how do I get it all out before I start to PC?

As the LC cools, the absorption rate of the water decreases.  That is why you are seeing sediment the next day.  I do not claim to be an expert on this and have only gotten 2 LC's to work so far and have tried many.  I would re-filter with 2 coffee filters and re-PC, allow it to cool and see if any sediment forms.  If so, repeat the process until no sediment.  You can use this information to systematically reduce the amount of LME/Dex you use at the start until you have a formula that works for you that will not produce sediment and will viable after 1 round of PC.

Sediment is fine, the myc loves it.

But LC from spores is a gamble, get a pack of agar for $1.69 and win big.

Can you make a liquid culture from agar then?  I thought you could only make it from spores...

Drop a wedge of agar into LC, bam, LC from agar. You can make LC from anything, a peice of grain, cake, mushroom, agar.

The PC makes more sediment, since it makes it above boiling temps, more crystalization. But yeah, shouldnt matter.
