i've come to the realization that i need more substrate to spawn to bulk.  i'm preparing for my first bulk grow, and i have (3) 1 pound bags of rye berry substrate that is 40 percent colonized, 15 pounds of straw, and 15 pounds of poo.  i want to do 50/50 poo/straw bulk and spawn to that, so i've ordered more rye berry mycobags.

NOW, my question.  i want to do a g2g transfer.  what's my best bet?  should  i buy some jars, sterilize them, and combing colonized grains with uncolonized grains?  or simply open the new bags and dump some colonized grains in the bags?

I would not go g2g with pre ordered spawn bags. just inject them like you did the last time. If the first 3 bags are done first that is fine just let them keep colonizing.

If your getting ready to go bulk I would wait until your ready and add a little bit from the bag to a jar of grain. It's not real sterile, but will work.
I did a G2G using a bag to jars and it was a pita, but doable in a GB.
Once you get a clean jar doing G2G in a glove box is'nt so bad.

deantheking, i'm trying to avoid getting more spores.

say i sterilize the jars, wipe down the grain bags with alcohol, and sterilize a knife or scissors.....can i just cut the bag, pour uncolonized grain in jar, followed by some colonized, and seal?  obviously i'm going to want to dress down in 'scrubs,' and sanitize the room (which is completely empty and has hard floors), but is this the way to go?

That approach, in open air is going to be highly prone to contamination. The only reasonable way to pull something like that off is in a flow hood. If you are thinking about this approach to save on spores is it possible for you to start a liquid culture while you are waiting for the new bags to come in? Do you have any spore solution left at all?

liquid culture is a method of cultivating mycelium in a rich liquid broth- normally a sugar, honey, or light malt extract (there are many recipes on here). Once these solutions are sterilized you can put a very small amount of sterile spore solution or sterile myc into it. This will give the myc a rich media to grow rapidly in. Once expanded this can be aspirated into a sterile syringe and used to inoculate BRF cakes, grains etc. The advantage to using liquid culture is that the myc tends to be very vigorous and when you inoculate a substrate it will tend to colonize much faster than spores. The potential downside is that liquid culture can sometimes be a receptor for contamination if your sterile technique isn't great.

Actually I think grain liquid culture might be the right route to go in this case.

g2g is possible as you describe, but a pain in the ass in a glovebox.

The cleaner solution is to do as guy above said and let the bags colonize and then make a glc to noc up more jars.

But generally yes everything you are suggesting would work.

edit: guy above is now talking about sugar lc...don't go that route. Just let the bag colonize, break it up, inject water, extract grain liquid culture. Lookitup in the teks "grain liquid culture" or "GLC".
