Cloning Fresh Tissue to GLC Jar?

I am about to attempt to cut some tissue from the inside of a fresh mushroom stem into a pint jar of some PC'd rye and im gonna use this tek http://www.shroomery.org/forums/showflat.php/Number/6817701/fpart/1/vc/1 and let colonize to make some GLC from. My question is...its a 2nd flush and the fruits are smaller than the 1st...will this make a difference in the GLC i make? Should i have cloned from the 1st flush? Will all the jars i inoc from this GLC produce small 2nd flush like fruits?

I just did this yesterday. It was my first time and hopefully I managed it contam free.  AFAIK you should clone your best fruits but I think you'll still get a mix of genetics in the sample.  Small fruit size (again from what I've read) can be due to lack of water in the substrate so might not be genetic (esp if your first flush fruits were bigger).

This is just my 2c - I'm not an experienced grower. Hopefully someone else can give you a more definitive answer.

The flush doesn't really matter, the genetics will be unarguably the same so there will be no difference in which mushroom you use. Variation in size, shape, and potency are more dependent on light, air, and moisture in the current environment that that specific mushroom is in.

I heard from RR that potency is about genetics. Not nutrients or fruiting conditions etc. So going for the biggest ones wouldn't make a diff? I did kinda forget about fanning/misting this grow during the later stages of its 2nd flush. And the fruits are smaller. But i always heard to go for the biggest most potent beefiest shroom when cloning. I gotta go to a friends house to get my jars and i gotta be back before 12pm to boil some rye i got soaking to do this hopefully later tonight when the PC cools down.

Fruit size is wildly variable depending on growing parameters. There is some small genetic component, but practically insignificant. You can grow any cube strain to giant proportions by playing with the substrate and growing parameters.

When cloning the tissue...do i have to flame sterilize the blade(s) before touching the stem to cut open and go for the tissue or is just wiping a shard blade with iso good enough?

RR makes a big point of it in his videos - always flame sterilize is the message I think :shocked:

Edit: The video also says to tear open the fruitbody rather than cut.

Isopropyl does not sterilize.

I lacked a scalpel but really wanted to clone the fruit so I just used a very sharp paring knife.  I wiped it with iso then flamed it with a tubolighter. Had a look today and the cloned bit seems to be fluffing up into myc and reaching for the grain! :awesome:
