Dionized/disstiled water for agar

Has anyone noticed a difference in using either for making agar. I used deionized water myself because that is what is in the lab i use. Could it affect growth, and does this leave room form more additives?

I used to use all sorts of expensive water purchased in plastic jugs which contribute to the huge waste problem.  I switched to tap water years ago and saw no decline in performance.  When I moved to my current location, I started using my well water, which is extremely hard and pH 9.  It also works very well.

The bottom line is use what is already available in your house.  If you're worried about it, boil first, then mix your agar.

^ What RR said... I bought distilled water initially for my agar/syringes as well, but quickly just gave up after seeing his advice.

At college, we use DI water for making prepping agar. At home, I use tap.

In my experience, the dissolved minerals in tap/well water can ONLY benefit the diversity of the nutrient profile of your agar.

HOWEVER, I must include a tad of anecdotal evidence.....

After a snow/icing up here in Ohio, they heavily salt the roads, and this leeches into our well at times.  THAT is the ONLY time I have ever noticed any decrease of mycological performance from just normal tap water. (I would think due to the overwhelming amt. of salts present.)

Do you know why?  I would guess its simply for repeatability as they can exclude water variability from being a signifigant factor in the agar's charecteristics.  I would think if anything it would help when making media, but I guess it depends on what your after in your agar.

In chem labs, it's to reduce/prevent contamination *the more chemistry related kind of contams, not the bacterial kind*
Would suck if trace amounts of minerals in the tap screwed over a carefully prepared experiment.
In bio labs, DI water to make defined medium & since tap water tends to be chlorinated & have trace minerals & unknown byproducts, could affect results when growing bacteria.

Who knows what these might do to your experiment. o_o'
